沙参麦冬汤对放射性肺炎大鼠血浆IL-6,TNF-α,TGF-β_1的影响

摘　　要：目的:观察沙参麦冬汤对放射性肺炎大鼠血浆白介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumour necrosis factor-α,TNF-α)、转化生长因子-β1(transforming growth factor-β1,TGF-β1)含量的变化,评价沙参麦冬汤对放射性肺炎的防治效果。方法:将141只清洁级雄性SD大鼠随机分为正常对照组、模型组、沙参麦冬汤预防组(中预组,10.3 g·kg-1)、沙参麦冬汤治疗组(中治组,10.3 g·kg-1)、地塞米松治疗组(西治组,10.4×10-3g·kg-1)。除正常对照组外,其余各组均给予造模。各组药物均用生理盐水稀释成等容积。除中预组于照射前1周起开始ig沙参麦冬汤外,其余各组于造模后第2天起,正常对照组、模型组ig等容积的生理盐水,中治组ig沙参麦冬汤,西治组ig地塞米松混悬液,每日1次,直至照射后第6周。大鼠饲养期间每周称体重1次,每周按体重变化调整1次剂量。于照射后2,4,6周,正常对照组取7只,其余各组随机取10只大鼠。10%水合氯醛溶液ip麻醉大鼠,心脏采血约5 mL,置肝素抗凝管中,4℃3 000 r·min-1离心10 min,分离血浆,置-20℃冰箱保存待查。采用ELISA法检测血浆中TGF-β1,TNF-α,IL-6的含量。结果:与正常对照组比较,2,4,6周模型组大鼠血浆中IL-6,TNF-α,TGF-β1含量均有显著升高(P<0.01)。与模型组比较,2,4,6周中预组、中治组、西治组大鼠血浆中IL-6,TNF-α,TGF-β1显著降低(P<0.01或P<0.05)。中预组低于中治组、西治组(P<0.01),中治组低于西治组(P<0.01)。结论:沙参麦冬汤可能通过降低血浆中IL-6,TNF-α,TGF-β1含量,抑制放射性肺炎肺泡炎性反应,减轻肺纤维化进程。Objective：To evaluate the prevention and treatment action of Shashen Maidong decoction and thus explore the possible mechanisms.Method：One hundred forty-one SD rats were randomly divided into five groups,control group,model group,Shashen Maidong decoction 10.3 g · kg-1 prevention group,Shashen Maidong decoction 10.3 g · kg-1 treatment group,and dexamethasone 10.4 mg · kg-1 treatment group.The rats model of radiation pneumonitis was established by exposeing rats to the chest X-ray except control group.The prevention group was given Shashen Maidong decoction one week before modeling,while the corresponding drug solutions of other four groups were administrated from the second day after modeling respectively once per day.Rats were anesthetized by intraperitoneal injecting 10％ chloral hydrate.The blood was collected.The levels of plasma interleukin-6 （IL-6）,transforming growth factor-β1 （TGF-β1） and tumour necrosis factor-α （TNF-α） were measured by ELISA at 2,4,6 weeks seperately after modeling.Result：Compared with control group,IL-6,TGF-β1 and TNF-α increased in model group （P ＜ 0.01） at 2,4,6 weeks.Compared with model group,the levels of serum IL-6,TGF-β1,TNF-α in Shashen Maidong decoction prevention group,Shashen Maidong decoction group and dexamethasone treatment group were significantly lowered （P ＜ 0.01,or P ＜ 0.05）.The levels of IL-6,TGF-β1,TNF-α in Shashen Maidong decoction prevention group were lower than Shashen Maidong decoction group and dexamethasone treatment group （P ＜ 0.01）,and in dexamethasone treatment group which was higher than that of Shashen Maidong decoction treatment group （P ＜ 0.01）.Conclusion：The Shashen Maidong decoction may inhibit the inflammation of alveolar and improve the process of pulmonary fibrosis in radiation peumonitis by reducing the levels of IL-6,TGF-β1,TNF-α.
