补肾化痰方对去卵巢大鼠骨组织TNF-α和IL-6的影响

摘　　要：目的:观察补肾化痰方对去卵巢骨质疏松症大鼠骨组织肿瘤坏死因子-α(TNF-α)和白细胞介素-6(IL-6)表达的影响,探讨补肾化痰方对去卵巢大鼠骨代谢影响的可能机制。方法:SD大鼠随机分为5组:模型组(A)、假手术组(B)、利维爱组(C)、补肾化痰方低剂量组(D)、补肾化痰方高剂量组(E)。行双侧卵巢切除术后3月,C,D,E组分别以利维爱0.23 mg·kg-1、补肾化痰方9.4 g·kg-1、补肾化痰方18.8 g·kg-1灌胃给药,A,B组予以等体积的生理盐水。1次/d,给药时间为8周。术后5个月,双能X线骨密度测定仪检测大鼠股骨骨密度,免疫组化检测大鼠骨组织TNF-α和IL-6表达。结果:补肾化痰方低、高剂量组均能显著提高去卵巢骨质疏松症大鼠的骨密度,增加骨小梁数量和密度,降低骨组织TNF-α和IL-6表达水平。结论:补肾化痰方对去卵巢大鼠骨质疏松症有较好的治疗作用,其作用机制与降低骨组织中细胞因子TNF-α和IL-6水平有关。Objective：To observe the influence of Bushen Huatan decoction on tumor necrosis factor （TNF-α） and interleukin-6 （IL-6） in ovariectomized rats and to explore the therapeutic mechanism of the drugs.Method：Female SD rats were divided into five groups：model group （A）,sham operation group （B）,livial group （C）,Bushen Huatan decoction low dose group （D） and high dose group （E）.Except for group B,all rats were ovariectomized.Drug treatment began at three months after the operation.Group C was treated with livial at the dose 0.23 mg ·kg-1,group D and E were treated with the low dose 9.4 g ·kg-1 the large dose 18.8 g ·kg-1 and Bushen Huatan decoction respectively A,B group recewed the same volume of physiological saline.After 8 weeks intragastric administration by once a day,the rat＇s bone mineral density was measured by dual energy X-ray absorptionmetry.Levels of TNF-α and IL-6 in bone tissue were determined by using immunohistoehemistry techniques.Result：Bushen Huatan decoction can evidently increase bone density as well as the trabecular bone volume and density,and decrease the levels of TNF-α and IL-6 in bone.Conclusion：Bushen Huatan decoction can effectly prevent osteoporosis in female rats induced by ovariectomy,and the mechanism maybe lowing levels of TNF-α and IL-6 in bone tissue.
