摘 要:目的观察壮医针挑疗法对哮喘模型小鼠肺组织miRNA-126和血清IL-4、IFNl及IgE的影响,探讨其起效的可能机制。方法将30只6~8周龄SPF级雌性BALB/c小鼠随机分为对照组、模型组、针挑组,每组10只。模型组、针挑组采用卵清蛋白致敏及激发制备哮喘小鼠模型,针挑组在此基础上采用壮医针挑疗法,针挑大椎、肺俞、定喘、风门、肾俞、脾俞,每日1次,7 d为1个疗程。疗程结束后取材,RT-PCR法检测肺组织miRNA-126表达,ELISA法检测血清IL-4、IFN-1,及IgE水平。结果模型组miRNA-126表达水平显著高于对照组(P〈0.01)。针挑组miRNA-126表达水平显著低于模型组(P〈0.01)。与对照组比较,模型组血清IL-4和IgE水平均显著升高(P〈0.01),而IFN-y水平显著降低(P〈0.01)。与模型组比较,针挑组血清IL-4和IgE水平均显著降低(P〈0.01),而IFN-y水平显著升高(P〈0.01)。结论 壮医针挑疗法能够下调肺组织miRNA-126表达,降低血清IL-4及IgE水平,上调血清IFN-y水平,恢复IL-4、IFN-Y水平,纠正Thl/Th2失衡状态可能是其起效环节之一。Objective To investigate the effect of Zhuang nationality medical picking therapy on lung miRNA-126 and serumIL-4, IFN-y and IgE in a mouse model of asthma and explore the possible mechanism by which it produces the effect on asthma.Method Thirty female SPF BALB/c mice aged 6-8 weeks were randomly allocated to control, model and picking therapy groups,10 mice each. A mouse model of asthma was made by ovalbumin sensitization and challenge in the model and picking therapygroups. In addition, the picking therapy group received needle picking out treatment at Dazhui (GV14), Feishu (BL13), Dingehuan(Ex-B 1), Fengmen (BL12), Shenshu (BL23) and Pishu (BL20) once daily, seven times as a course. The materials were taken after theend of treatment course. Lung expression of miRNA-126 was examined by RT-PCR. Serum IL-4, IFN-7 and IgE levelswere measured by EL1SA. Result The expression of miRNA-126 was significantly higher in the model group than in the controlgroup (P〈0.01) and significantly lower in the picking therapy group than in the model group (P〈0.01). Compared with the controlgroup, serum IL-4 and IgE levels increased significantly and IFN-y levels decreased significantly in the model group (both P〈0.01).Compared with the model group, serum IL-4 and IgE levels decreased significantly and IFN-7 levels increased significantly in thepicking therapy group (both P〈0.01). Conclusion Zhuang nationality medical picking therapy can down-regnlate lung expressionof miRNA-126, decrease serum IL-4 and IgE levels, up-regulate serum IFN-y levels and restore IL-4 and IFN-y levels. Correction ofThl/Th2 imbalance may be one of the links via which it produces the effect.
