摘 要:目的:初步探讨饱和和不饱和长链脂肪酸通过G蛋白受体120(GPR120)对脂肪细胞炎症、内质网应激及胰岛素信号分子的影响及其机制。方法:选取5例择期行胆结石胆囊手术的患者的大网膜脂肪组织,分离成熟脂肪细胞并进行培养。分别进行长链多不饱和脂肪酸DHA研究和长链饱和脂肪酸PA研究。应用实时定量PCR法和western blot法检测GPR120、FABP4、肿瘤坏死因子α(TNFα)、内质网应激蛋白p-eIF2α及胰岛素信号通路蛋白IRS-1的基因和蛋白表达水平。结果:(1)DHA刺激下,GPR120和FABP4的表达均显著增加(均P<0.01),同时,TNFα(P<0.01)和p-eIF2α(P<0.05)表达下调,IRS1(P<0.05)表达上调。加入GPR120特异性抑制剂GW9508后,GPR120和FABP4的表达均显著减少(均P<0.01),TNFα(P<0.01)和p-eIF2α(P<0.01)表达较不加GW9508时上调,IRS1(P<0.05)表达下调。(2)在PA刺激下,除TNFα(P<0.05)和p-eIF2α(P<0.01)表达上调、IRS1(P<0.05)表达下调外,其余结果同(1)。结论:GPR120介导了脂肪酸诱导的胰岛素信号通路的调控,并在饱和长链脂肪酸和不饱和长链脂肪酸的作用中起到平衡调控作用。Objective:To explore saturated and unsaturated long-chain fatty acid on the adipocyte inflammation,endoplasmic reticulum (ER)stress and insulin signaling moleculars through G protein receptor 120 (GPR120)and it's related mechanism.Methods:Greater omentum adipose tissue were selected from five selective gallstone surgical patients to separate and culture the mature adipocytes.Research on DHA and PA were performed.Real time PCR and western blot were used to evaluate the gene and protein expression of GPR120,FABP4,TNFot,p-eIF2α and the insulin signaling protein IRS-1.Results: (1)Under the stimulation of DHA,the expression of GPR120(P〈0.01)and FABP4(P〈0.01)increased,the expression of TNFα(P〈0.01)and p- eIF2ct (P〈0.05)decreased,IRS1 (P〈0.05)increased.But after added GPR120 specific inhibitor GW9508,the ex- pression of GPR120(P〈0.01 )and FABP4(P〈0.01 )decreased,TNFα (P〈0.01)and p-eIF2α(P〈0.01 )increased,IRS1 (P〈O.05)expression decreased.(2)Under the stimulation of PA,exeept the increased expression of TNFα (P〈 0.05),p-eIF2α(P〈0.01)and the decreased expression of IRSl(P〈0.05),the results was same as DHA.Conclusion:GPR120 mediates the control of fatty acid-induced insulin signaling pathway,balances the function of saturated and unsaturated fatty acid.
