麝香乌龙丸对兔膝骨关节炎软骨组织中IGF-Ⅰ及Ⅱ型胶原表达的影响

摘　　要：目的:通过研究兔膝骨关节炎(KOA)膝关节软骨中IGF-Ⅰ(胰岛素样生长因子)及Ⅱ型胶原在软骨细胞的表达变化,探讨麝香乌龙丸对关节软骨的修复及保护作用是否与调节IGF-Ⅰ及Ⅱ型胶原在软骨细胞中的表达有关。方法:40只健康日本大耳白兔,随机分成4组,即正常组、模型组、麝香乌龙丸组、抗骨增生片组。除正常组外,其余三组用1.6%木瓜蛋白酶膝关节腔内注射建立KOA模型,于第三次注射木瓜蛋白酶后第2天开始给药。给予麝香乌龙丸组麝香乌龙丸0.6g/kg-1,抗骨增生片组抗骨增生片0.36g/kg-1(生理盐水混悬,相当于成人剂量的6倍)灌胃,正常组与模型组灌入等量生理盐水,4周后检测关节软骨中IGF-Ⅰ、Ⅱ型胶原的水平。结果:与正常组比较,模型组IGF-Ⅰ阳性表达明显减低,Ⅱ型胶原阳性表达明显升高;与模型组比较,麝香乌龙丸组与抗骨增生片组IGF-Ⅰ阳性表达均增高,软骨Ⅱ型胶原阳性均降低;IGF-Ⅰ在麝香乌龙丸组阳性表达高于抗骨增生片组(P<0.05),Ⅱ型胶原在麝香乌龙丸组阳性表达低于抗骨增生片组(P<0.05)。结论:麝香乌龙丸可以通过调节IGF-Ⅰ及Ⅱ型胶原在软骨细胞中的表达,对关节软骨起到修复和保护作用。Objective： Research on the expression of IGF-Ⅰ （IGF） and type Ⅱ collagen in chondrocytes of the rabbit of knee osteoarthritis （KOA）, and discuss the possible mechanism of Shexiang Wulong Pill on articular cartilage repairment and protection effect. Methods： 40 healthy male rabbits （weight 2. 5±0. 15kg） were randomly divided into four groups： the normal group, model group, Shexiang Wulong Pill group, Kanggu Zengsheng Tablet group. KOA models were established by injection of 1. 6% papain to the rabbit knee joint cavity. The drugs were administrated on the second day after papain injection at the third time. Rabbits in Shexiang Wulong Pill group were administrated by ig with Shexiang Wulong Pill in dose of 0. 6g/kg^-1 , Kanggu Zengsheng Tablet group by ig with Kanggu Zengsheng tablet in dose of 0. 36g/kg^-1 （saline suspension, equivalent to 6 times of adult doses respectively）. While the rabbits in the nomal and the model groups were treated with nomal saline. 4 weeks later the IGF-Ⅰ , Ⅱ type collagen levels were examined in the articular cartilage. Results： Levels of IGF- Ⅰ , Ⅱ collagen in cartilage were significantly lower in the model group, compared with that in the nomal group （ P 〈0. 05） ; compared with that in the model group, the levels of IGF-Ⅰ and Ⅱ collagen in cartilage were significantly higher in the Shexiang Wulong Pill group and the Kanggu Zengsheng tablet group which had no significant difference between theses two gorups. Conclusion： Shexiang Wulong Pill can regulate the expression of IGF-Ⅰ and type Ⅱ collagen in chondrocytes of articular cartilage and have repare and protective function on articular cartilage.
