摘 要:目的:观察丹红注射液对大鼠局灶脑缺血模型后外周血内皮祖细胞数量及神经功能缺损的影响。方法:SD大鼠按随机数字表法分为假手术组、脑梗死组(对照组)和脑梗死丹红注射液治疗组(丹红治疗组)。对照组和丹红治疗组采用线栓法建立左侧永久性大脑中动脉栓塞(permanent Middle cerebral artery occlusion,p-MCAO)模型。术后24 h±2 h、72 h±2 h、168 h±2 h抽取尾静脉血,通过流式细胞仪测定CD133+/VEGFR-2+双阳性细胞数量并进行神经功能缺损评分。结果:3组术前外周血CD133+/VEGFR-2+双阳性细胞数比较差异无统计学意义。丹红治疗组大鼠术后24 h、72 h、168 h外周血CD133+/VEGFR-2+双阳性细胞数百分率较对照组升高(64.60%(14.62%,82.32%)vs 1.38%(0.05%,24.14%),24.36%(7.38%,72.36%)vs 2.78%(0.09%,32.17%),4.53%(0.57%,11.16%)vs 0.46%(0.07%,0.98%),P<0.01)。丹红治疗组大鼠术后24 h、72 h、168 h神经功能评分较对照组低(2(1,4)vs 2.5(0,4),1(0,4)vs 2(0,4),0(0,1)vs 3(0,4))。结论:丹红注射液有可能提高脑梗死后外周血内皮祖细胞数目,具有改善神经功能缺损的作用。Objective: To investigate the influence of Danhong Injection on the quantity of endothelial progenitor cells in peripheral blood and the neurological functional score in rats after Focal Cerebral Ischemia. Methods: Male SD rats were divided randomly into sham-operated group, ischemia (operated) group and Danhong (ischemia treated with Danhong) group. Operation group and Danhong group were established into the permanent Middle Cerebral Artery Occlusion (p- MCAO) model. The neurological function deficit was evaluated at 2, 24 ± 2, 72 ± 2, 168 ± 2 h after operation. The quantity of CD133 +/VEGFR-2 + dual-positive cells were determined by the flow cytometer at 24 ± 2, 72 ± 2, 168± 2 h after operation, and statistically analyzed. Results The quantity of CD133 +/VEGFR-2 + dual-positive cells at 24h, 72 h and 168h in Danhong group was significantly increased than operation group (64. 60% (14.62% , 82. 32% ) vs 1.38% (0.05%, 24.14%),24.36% (7.38%, 72.36%) vs 2.78% (0.09%, 32.17%) , 4.53% (0.57%, 11.16%) vs 0.46% (0.07% , 0.98% ) , P 〈 0.01 ) ). The neurological functional scores were improved in Danhong group than operation group at24h, 72 h and 168h (2 (1, 4) vs2.5 (0, 4), 1 (0, 4) vs2 (0, 4), 0 (0, 1) vs3 (0, 4)). Conclusion : Danhong Injection may increase the number of endothelial progenitor cells and improve neurological impairment after brain ischemia.
