摘  要:为了研究不同鸭种胚胎期和出雏早期骨骼肌(胸肌和腿肌)NFAT5基因mRNA表达规律及其与生长发育的相关性,本试验克隆了鸭NFAT5基因cDNA部分片段,并运用荧光绝对定量PCR技术检测了生长速度差异较大的高邮鸭和金定鸭13、17、21、25和27胚龄及7日龄胸肌和腿肌中NFAT5 mRNA的表达水平。结果显示,胸肌和腿肌NFAT5 mRNA在2种鸭中均具有相似的表达模式,均在13胚龄时表达量达最高,且均显著高于其他各胚龄和7日龄(P<0.05),而出雏后7日龄的表达水平显著高于27胚龄(P<0.05)。相关性分析结果显示,2种鸭胸肌和腿肌NFAT5 mRNA表达量与其胸肌质量、腿肌质量和体质量均呈极显著负相关(P<0.01),而与组织中IGF-I mRNA的表达量呈极显著正相关(P<0.01)。表明鸭胸肌和腿肌NFAT5与IGF-I存在协同效应,两者可能共同参与了对骨骼肌生长发育的调节。Partial sequences of NFAT5 cDNA in two duck breeds, Gaoyou duck and Jinding duck were cloned, and the mRNA expression levels in the pectorale and leg muscles were detected by quantitative RT-PCR at 13-, 17-, 21-, 25-, 27-day embryonic age and at seven days of age. The results showed that the NFAT5 mRNA expressions in the pectorale and leg muscles of Gaoyou duck and Jinding duck were similar, with the highest value at 13 embryonic age. At severn day, the NFAT5 mRNA expression of 7-year-old ducks was significantly higher than that of ducks at 27-day embryonic age ( P〈0. 05 ) . The expression levels of NFAT5 in the pectorals and legs of two duck breeds were negatively correlated with the pectoral weights, leg weights, and&amp;nbsp;body weights, while positively correlated with IGF-I mRNA expression ( P〈0. 01 ) . These results suggested that there was a synergism between NFAT and IGF-I which may be involved in the regulation of growth and development of duck skeletal muscle.
