摘  要:在配制H9亚型禽流感疫苗过程中添加免疫增强剂VA5制成VA5-H9疫苗免疫鸡,或将VA5制备成油乳剂以疫苗伴侣形式与商品化H5亚型禽流感疫苗混合(VA5-H5)后免疫鸡,监测鸡的抗体持续期、疫苗安全性和鸡淋巴细胞的转化情况。抗体检测结果表明,VA5与H5或H9亚型禽流感疫苗混用能有效延长抗体持续期4周,H5抗体提前1周达到7lg2。安全性研究表明,以VA5-H9疫苗经一次单剂量、一次2倍剂量和单剂量间隔2周重复免疫这3种方式免疫鸡后,含VA5的疫苗与常规疫苗的吸收类似,说明VA5不影响鸡体对疫苗的吸收。淋巴细胞转化试验结果显示,VA5能提高免疫鸡淋巴细胞转化效率。可知,获得的VA5免疫增强剂能有效延长抗体持续期,且不影响疫苗的安全性,并提高免疫鸡的淋巴细胞转化活性。The immunopotentiator VA5 was added to the H9 subtype avian influenza ( AI ) vaccine during its preparation processes, and the H5 subtype AI vaccine was mixed with chaperon form of VA5 prepared as oil emulsion. Chickens were inoculated with the VA5-contained or -free vaccines for immunization, and the hemagglutinin inhibition ( HI) titers of chicken sera against corresponding antigen were measured. Three different regimes of vaccine were ad-ministered for VA5 safety test, including single injection with single dose, single injection with double doses or re-peated single-dose injection at 2-week interval. The role of VA5 in chicken lymphocytes transformation was also investigated. The antibody persistence period of chickens in either H5 or H9 VA5-contained vaccine group were prolonged by 4 weeks than that in VA5-free vaccine group, and the antibody titer against H5 antigen was 1 week earlier to reach 7lg2 in chickens immu-nized with VA5-contained vaccine. The similar absorption behavoirs of the vaccines administered in three different regimes by chickens revealed that the absorption is independent of VA5. Stimulation index indicated that VA5 improved the efficiency of lymphocyte transformation in vaccinated chicken. In sum, the VA5 immunopotentiator is effective in prolonging the antibody persistence period without compromise the safety of vaccine and improving lymphocyte transformation.
