摘  要:氮素营养对蔬菜产量和品质起着关键作用。为了鉴定不同氮源以及氮源不同浓度和作用时间对不结球白菜氮代谢的影响,采用实时定量PCR法分别检测不结球白菜根系和叶片中的氮代谢关键酶基因(硝酸还原酶基因BcNR、亚硝酸还原酶基因BcNiR、谷氨酰胺合成酶基因BcGS1和BcGS2)的表达。结果显示:硝态氮处理有利于4种基因的表达,30 mmol/L的硝态氮处理时白菜根系和叶片中的BcNR与BcNiR的表达量达到最大,20 mmol/L硝态氮处理时BcGS1和BcGS2基因表达量达到最大;硝态氮诱导初期,4种基因的表达水平均显著提高,后期呈下降趋势,30 mmol/L的硝态氮处理下4~6 h BcNR与BcNiR的表达量最大,6 h时BcGS1基因表达量最大,诱导8 h时BcGS2基因表达量最大;铵态氮可促进BcGS1和BcGS2基因的表达,但高浓度的铵态氮抑制了BcNR与BcNiR的表达。Nitrogen is the key factor affecting the yield and quality of vegetables. In order to identify the influence of ni-trogen sources, concentrations and application time of duration on nitrogen metabolism in non-heading Chinese cabbage (Brassica campestris ssp. chinensis), mRNA levels of the genes BcNR, BcNiR, BcGS1 and BcGS2(encoding nitrate reductase, nitrite re-ductase, cytoplasmic glutamine synthetase and chloroplast glutamine synthetase, respectively) related to nitrogen metabolism in the roots and leaves were detected by real time PCR. The maximum expressions of BcNR and BcNiR in roots and leaves were in-duced by 30 mmol/L NO3--N, while the maximum expressions of BcGS1and BcGS2 were induced by 20 mmol/L NO3--N. The expressions of four genes were significantly increased by NO3--N treatment at early stage, then declined. 30 mmol/L NO3--N in-duced the maximum expressions of BcNR and BcNiR in roots and leaves at 4-6 h, the maximum expression of BcGS1 at 6 h, and BcGS2 at 8 h. The expressions of BcGS1 and BcGS2 were enhanced by NH4+-N, but high level of NH4+-N treat-ment inhibited the expressions of BcNR and BcNiR.
