沉香镇痛有效部位及其物质基础研究

摘　　要：目的筛选沉香药材中发挥镇痛作用的有效部位及有效成分。方法采用加热回流法提取得到沉香醇提液,依次萃取得到其石油醚、二氯甲烷、乙酸乙酯、正丁醇各部位;运用热板法、醋酸致小鼠扭体法,考察各部位对小鼠镇痛作用的影响,筛选得到沉香镇痛有效部位;运用UPLC法分析沉香镇痛作用有效部位的入血成分,探讨其镇痛作用的物质基础。结果沉香醇提液及其石油醚部位对热板法引起的小鼠疼痛有显著抑制作用(P<0.05);沉香醇提液的正丁醇萃取部位对醋酸引起的小鼠疼痛有显著的抑制作用(P<0.05),所对应的成分为Aquilarone E。结论沉香醇提液的石油醚部位和正丁醇部位是沉香镇痛作用的有效部位;正丁醇部位发挥作用的物质可能是Aquilarone E。Objective To study the analgesic effective parts and active substance of Aquilaria sinensis （Lour.） Gilg. Methods Use heating reflux method to get the alcohol extract （AE） of Aquilaria sinensis （ Lour. ） Gilg, and then extracted with petroleum ether, dichloromethane , EtOAc and n- BuOH to get petroleum ether extract（ PE）, dichloromethane extract（ DE）, EtO- Ac extract（EAE） and n - BuOH extract（BE）. The writhing responses induced by acetic acid and the hot - plate test were used to evaluate the analgesic activity of different extracts in mice. The UPLC was used to analysis the active substance of analgesic effective parts. Results The result is that the AE and the PE revealed analgesic activity in the hot - plate test. The BE revealed analgesic activity in writhing response induced by acetic acid, the active compound may be Aquilarone E. Conclusion PE and BE are the effective parts of Aquilaria sinensis （ Lour. ） Gilg with analgesic activity. The active substance of BE may be Aquilarone E.
