摘  要:目的利用响应面分析法对番石榴叶中总黄酮的超声辅助提取条件进行研究。方法以总黄酮的提取率为指标,在单因素试验的基础上,采用响应面设计法考察乙醇体积分数、液料比、超声功率对总黄酮提取条件的影响。结果番石榴叶中总黄酮的最佳提取工艺条件为:75%(体积分数)乙醇,液料比60∶1(mL∶g),超声功率150 W,提取时间30 min,提取温度50℃,提取2次。在此条件下,番石榴叶总黄酮的提取率为0.497%,与理论值(0.500%)接近。结论在响应面分析法优选出来的最佳条件下,总黄酮的实际提取率与理论提取率非常接近,说明通过响应面分析法优化番石榴叶中总黄酮的提取工艺条件可行。Objective To study on the effects of different factors on the extraction of total flavonoids from leaves of Psidium guajava and optimize the ultrasound assisted extraction of total flavonoids by using response surface methodology. Methods Ethanol concentration, ratio of material to liquid and ultrasonic power were evaluated basing on single factor experiments with the yields of total flavonoids as index. Then response surface methodology with three-factors and three-levels was carried out for optimizing the extraction process of total flavonoids. Results The optimum conditions were as follows:extracting with 75%ethanol for twice and 30 minutes each time at a temperature of 50 ℃,liquid-solid ratio of 60∶1(mL∶g), and ultrasonic power of 150 W. Conclusion The actual yields of total flavonoids were very close to the predicted values,indicating that response surface analysis used in this experiment was feasible.
