摘 要:目的探讨柴芩承气汤(CQCQD)对急性坏死性胰腺炎大鼠肠道平滑肌细胞三磷酸肌醇浓度的影响。方法将30只SD大鼠随机分成对照组、急性坏死性胰腺炎(ANP)组及CQCQD组,采用8%左旋精氨酸腹腔注射制作AP大鼠模型。CQCQD组管喂CQCQD,于6h取血清检测乙酰胆碱(Ach)、胆囊收缩素-8(CCK-8)及5-羟基色氨酸(5-HTP)水平,胰腺组织做病理检查,结肠检测平滑肌细胞内三磷酸肌醇(IP3)及Ca2+浓度变化。结果CQCQD组胰腺病理评分(4±0.7)低于ANP组(7±1.1)(P<0.05)。CQCQD组血清CCK-8浓度(31.5±6.2)ng/mL低于对照组(44.0±8.1)ng/mL和ANP组(46.0±7.6)ng/mL(P<0.05);ANP组血清Ach浓度(236.1±23.6)ng/mL低于对照组(310.0±26.1)ng/mL和CQCQD组(298.5±24.7)ng/mL(P<0.05);ANP组肠道平滑肌细胞IP3(16.3±1.1)pg/mL及钙离子浓度([Ca2+]i)(108.0±15.5)低于对照组肠道平滑肌细胞IP3(21.9±1.3)pg/mL及[Ca2+]i(141.2±20.2)和CQCQD组肠道平滑肌细胞IP3(21.6±1.7)pg/mL及[Ca2+]i(138.8±16.3)(P<0.05)。结论 ANP存在血清Ach及平滑肌细胞内IP3水平降低,CQCQD可促进ANP大鼠血清Ach水平及平滑肌细胞IP3浓度升高,促进肠道平滑肌细胞钙库释放[Ca2+]i,但具体作用机制尚需进一步研究。Objective To investigate the effect of Chai-Qin-Cheng-Qi Decoction(CQCQD)on Inositol triphosphate(IP3)concentration of intestinal smooth muscle cells in rat with acute necrotizing pancreatitis(ANP).Methods Thirty Sprague Dawley rats were randomized into control group,ANP group and CQCQD group(n=10×3).The AP model was induced by two intraperitoneal injections of 4g/kg L-Arginine at 1hapart,while rats in control group received saline injections.At 72 hafter the first injection of L-Arginine,rats in the treatment group were intragastrically administered 1mL/100 g CQCQD every 2hfor 3times,whilst rats in the other two groups received same amount of saline feeding.Rats were humanly killed 2hafter last feeding of either CQCQD or saline.The serum was collected for determination of Ach,CCK-8 and 5-HTP,pancreatic tissue was collected for pathological examination,colonic smooth muscle cells were collected for detection IP3 and Ca2+concentration.Results The histopathological score of pancreatic tissues in CQCQD group(4±0.7)was lower than those in the ANP group(7±1.1)(P〈0.05).The serum CCK-8in the CQCQD group(31.5±6.2)ng/mL was lower than those in the control group(44.0±8.1)ng/mL and ANP group(46.0±7.6)ng/mL(P〈0.05).The serum Ach in the ANP group(236.1±23.6)ng/mL was lower than those in the control group(310.0±26.1)ng/mL or CQCQD group(298.5±24.7)ng/mL(P〈0.05).The IP3(16.3±1.1)pg/mL and[Ca2+]i(108.0±15.5)of colonic smooth muscle cells in the ANP group was lower than those in the control group[(21.9±1.3)pg/mL,(141.2±20.2)]or CQCQD group[(21.6±1.7)pg/mL,(138.8±16.3)](P〈0.05).Conclusion CQCQD can promote serum levels of Ach and the IP3 concentration of smooth muscle cells of ANP to promote the release of[Ca2+]i on form intestinal calcium store,but the specific mechanism needs further study.
