摘 要:目的探讨血塞通胶囊对大鼠梗死脑组织自体神经干细胞增殖分化的影响。方法参照改良ZeaLonga法建立大鼠大脑中动脉缺血再灌注损伤模型,将造模成功大鼠随机分为模型组和中药高、中、低剂量组,每组40只,另设健康大鼠40只为正常组。中药高、中、低剂量组分别以血塞通胶囊60、40、20mg/(100g·d)灌胃,正常组和模型组以1ml/(100g·d)生理盐水灌胃,各组均灌胃28天。分别于给药后1、3、7、14、28天取材8只大鼠,检测梗死边缘区5-溴脱氧尿嘧啶核苷(BrdU)、微管相关蛋白-2(MAP-2)、胶质纤维酸性蛋白(GFAP)、3型β微管蛋白(IIIβ-Tubulin)表达变化。结果正常组偶见BrdU、MAP-2、GFAP、IIIβ-Tubulin表达阳性细胞,模型组和中药高、中、低剂量组各时间点梗死边缘区BrdU、MAP-2、GFAP、IIIβ-Tubulin表达阳性细胞均较正常组显著增加(P<0.05);中药高、中剂量组在给药后1、3、7、14天时BrdU、MAP-2、GFAP、IIIβ-Tubulin阳性细胞数较模型组同时间点显著增多(P<0.05)。结论血塞通胶囊可促进梗死脑组织神经干细胞增殖分化,参与损伤脑组织的修复和再生作用。Objective To study the effects of Xuesaitong Capsules on the repair and regeneration of autologous neural stem cells in infarcted brain tissue of rats. Methods The middle cerebral artery ischemia-reperfusion injury model was established with modified Zea-Longa method. The rat models were randomized into the model group and Xuesaitong Capsules high-dose, middle-dose and low-dose groups, with 40 in each. Forty healthy rats were selected as the normal group. Xuesaitong Capsules high-dose, middle-dose and low-dose groups were gavaged with 60 mg/ (100 g · d), 40mg/ ( 100g · d) and 20mg/ ( 100g · d) of Xuesaitong Capsules respectively. The normal group and model group were gavaged with same volume of normal saline. The intragastric administration lasted for 28 day. Eight rats were sacrificed on the 1 st, 3rd, 7th, 14th and 28th day after administration respectively to detect the expressions of 5-bromodeoxyuridine (Brdu), microtubule associated protein-2 (MAP-2), glial fibrillary acidic protein (GFAP) and Ⅲβ-Tubulin. Results The Brdu, MAP-2, GFAP and Ⅲβ-Tubulin positive ceils were occasionally seen in the normal group. Comparing with the normal group, the Brdu, MAP-2, GFAP and Ⅲβ-Tubulin positive cells were sig- nificantly increased in the model group and Xuesaitong Capsules high-dose, middle-dose and low-dose groups (P 〈 0. 05 ). Comparing with the model group at the same time point, the Brdu, MAP-2, GFAP and Ⅲβ-Tubulin positive cells were significantly increased in Xuesaitong Capsules high-dose, middle-dose and low-dose groups ( P 〈 0. 05 ). Conclusion Xuesaitong Capsules can promote the proliferation and differentiation of neural stem cells in infarcted brain tissue and improve the repair and regeneration of damaged brain tissue.
