棉油皂脚中棉籽糖的提取纯化及HPLC-ELSD含量测定

摘  要:从棉油皂脚中分离纯化棉籽糖,并进行准确的含量测定。样品经热水提取,Sevage法去蛋白、活性炭脱色,Sephadex LH-20柱层析后纯化后得到棉籽糖,采用Sugar-D色谱柱,流动相为75:25(v/v)的乙腈水混合液,使用蒸发光散射器进行分析检测。纯化后得到浅黄色的棉籽糖针状结晶,紫外光谱分析显示,在195 nm波长处有明显吸收峰,260、280 nm等处无吸收峰,说明被测物为多糖,且不含蛋白质;活性炭脱色后脱色率为90.54%;棉籽糖在5.02~40.16μg线性关系良好,平均回收率为96.62%,纯化后的棉籽糖含量显著提高。结果表明最佳提取工艺重现性较好;HPLC-ELSD法测定棉籽糖含量结果准确可靠。The process of extracting raffinose from cottonseed Soapstock has been established, and determined by HPLC-ELSD. A raffinose was isolated and purified by hot water, Sevage deproteination, activated carbon decolourization and Sephadex LH-20; the analysis was carried out on ELSD detector with with mobile phase of acetonitrile and water(95:5, v/v). The raffinose of light yellow needle-like crystals was obtained by purification, UV-spectrophotometer analysis indicated that raffinose showed a signal of polysaccharide at 195 nm and no signals of protein, nucleic acid at 260 and 280 nm; the decolourization rate is 90.54%, there is a good linear from 5.02 μg to 40.16 μg for raffinose, the average recoveries of which is 96.62%. After purification levels of raffinose significantly improved. The optimized extraction process indicates satisfied repeatability and the method for determining phytosterols by HPLC-ELSD is accurate and reliable.
