复方儿茶止泻霜对番泻叶致泻大鼠结肠水通道蛋白3表达的影响

摘　　要：目的:探讨复方儿茶止泻霜对番泻叶致泻大鼠模型结肠组织水通道蛋白3(AQP-3)表达的影响。方法:将50只SD大鼠随机分为空白组、阴性对照组、阳性对照组、高剂量组和低剂量组5组,每组10只。模型建立成功后取大鼠结肠组织,应用逆转录-聚合酶链反应(RT-PCR)和蛋白质Western blot印迹法检测各组动物结肠AQP-3的表达水平。结果:RT-PCR和Western blot结果显示一致,高剂量组、低剂量组结肠AQP-3 mRNA和蛋白表达水平与阴性对照组、空白组比较均增多,高剂量组增加显著(F=48.687,P<0.05);高剂量组与阳性对照组相比较,其差异无统计学意义。结论:复方儿茶止泻霜对番泻叶致泻大鼠结肠AQP-3蛋白的表达有上调作用,且呈一定剂量的依赖性。Objective：To investigate the effect of Compound Catechu Anti-diarrhea Ointment on the expression of aquaporin 3（AQP-3）of colonic mucosa of diarrhea rats.Methods：Fifty SD rats were divided into 5 groups,per group for 10 rats,including diarrhea model（DM）group,low dose-（L）,and high dose-（H）Water extraction solution of Compound Catechu groups,positive control drug（PC）group,and normal control（NC） group.The expression of AQP-3 protein of colonic mucosa in different groups was determined by RT-PCR and Western blot methods.Results：Western blot and RT-PCR results showed that the expressions of AQP-3 in the colon region of high/low dose group were increased significantly than DM and NC group,AQP-3 of H group were obviously increased than L group（F=48.687,P＜0.05）,but similar with the expressions of AQP-3 in the colon region of PC group.Conclusion：Compound Catechu Anti-diarrhea Ointment can increase the expression level of AQP-3 protein in the colon region in diarrhea rats and play a important role in the drug therapy.
