摘  要:目的:探讨化浊解毒方对胰岛素抵抗(IR)大鼠脂代谢的影响。方法:将模型建立成功的胰岛素抵抗大鼠随机分为化浊解毒方组、模型组、空白对照组,每组16只。化浊解毒方组予化浊解毒颗粒按生药3 g/(kg·d)灌胃给药,空白对照组、模型组灌服等体积生理盐水,各组连续做相应处理8周。8周后检测各组大鼠血清甘油三酯(TG)、血清总胆固醇(TC)、游离脂肪酸(FFA)、大鼠肝脏二酰基甘油酰基转移酶2(DGAT2)mRNA表达以及肝脏组织TG含量水平。结果:模型组大鼠血清甘油三酯(TG)、血清总胆固醇(TC)以及游离脂肪酸(FFA)明显高于空白对照组(P<0.01);化浊解毒方组大鼠血清甘油三酯(TG)、血清总胆固醇(TC)以及游离脂肪酸(FFA)显著低于模型组(P<0.01);化浊解毒方组DGAT2mRNA表达较模型组显著降低(P<0.01);化浊解毒方组大鼠肝脏组织TG含量水平明显低于模型组(P<0.01)。结论:化浊解毒方可改善胰岛素抵抗大鼠的脂代谢紊乱,其作用机制可能是通过下调DGAT2基因表达,进而影响TG合成及TG在肝脏组织的蓄积。Objective:To explore the effect of Huazhuo Jiedu Formula (Turbidity -transforming, Toxin -resolving Formula) for lipid disorders rats with insulin resistance (IR). Methods:The model was successfully established in the insulin resistance rats and they were randomly divided into Huazhuo Jiedu Formula group, model group, blank control group, with 16 rats in each group. Huazhuo Jiedu Formula group was given Huazhuo Jiedu Granule at 3 g/(kg/d) crude drug by intragastrie administration. Blank control group and model group were fed with an equal volume of saline. Each group acted continuously as stated above for 8 weeks. After 8 weeks, we detected the serum triglyceride (TG), serum total cholesterol (TC), free fatty acid (FFA), diaeylglycerol aeyltransferase 2 (DGAT2) mRNA expression and liver tissue TG content levels. Results : The serum triglyeeride (TG) , the serum levels of total cholesterol (TC) and free fatty acid (FFA) of rats in the model group were significantly higher than those in the blank control group (P 〈 0.01 ). The serum triglyceride (TG) , the serum levels of total cholesterol (TC) and free fatty acid (FFA) of rats in the Huazhuo Jiedu Formula group were significantly lower than those in the model group (P 〈0.01 ). The expression of DGAT2 mRNA in Huazhuo Jiedu Formula group was significantly decreased compared with that of the model group (P 〈 0.01 ). The TG content level in liver tissue of Huazhuo Jiedu Formula group was significantly lower than that in the model group (P 〈 0.01 ). Conclusion : Huazhuo Jiedu Formula can improve lipid disorders rats with insulin resistance. Its mechanism is probably to reduce the DGAT2 gene expression, thereby affecting TG synthesis and TG accumulation in the liver tissue.
