摘 要:目的:观察健脾降脂中药对脾失健运膏脂转输障碍大鼠肝脏胆固醇代谢相关基因表达的影响,探讨健脾降脂中药干预胆固醇代谢的作用机制。方法:60只大鼠随机分为空白对照组、模型组、健脾降脂治疗组。采用劳倦过度加饮食不节结合高脂饲料喂饲造模法,治疗组予健脾降脂中药日一次灌胃,空白对照组、模型组给予等体积的生理盐水灌胃。全自动生化分析仪检测血清胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平。油红O染色观察肝脏脂质沉积。采用实时定量RT-PCR和Western blotting技术检测大鼠肝组织B类Ⅰ型清道夫受体(SR-BⅠ)、肝低密度脂蛋白受体(LDL-R)、卵磷脂胆固醇酯酰基转移酶(LCAT)等基因mRNA和蛋白水平。结果:与空白对照组相比,模型组大鼠血清TC、LDL-C水平显著升高(P<0.05),HDL-C水平显著降低(P<0.05),肝细胞中见大量脂质沉积,肝脏LDL-R、SR-BⅠ、LCAT基因表达显著下调。与模型组相比,健脾降脂治疗组大鼠TC、LDL-C显著下降,HDL-C显著升高,肝细胞中脂质沉积明显减少,肝脏LDL-R、SR-BⅠ、LCAT基因表达显著上调。结论:健脾降脂中药具有调节血脂的作用,其作用机制可能与调控肝组织胆固醇代谢相关基因表达有关。Objective :To observe the effect of Jianpi Jiangzhi Recipe on expression of genes associated with cholesterol metabolism in the liver of the rats with dysfunction of the spleen in transportation, so as to illuminate the intervention mechanism of Jianpi Jiangzhi Recipe on cholesterol metabolism. Methods : Sixty rats were randomly divided into blank group, model group and treatment group with Jianpi Jiangzhi Recipe. Using overstrain method and diet not festival with high fat feedstuff feeding, treatment group were filled the stomach with Jianpi Jiangzhi Recipe once a day,while blank group and model group were filled the stomach with normal saline of the same volume. The levels of serum lipid were tested by biochemical methods. Oil red staining method was used to observe the liver structure and lipid sink. Real - time PCR and western blotting methods were used to test the changes of ex- pression of SR - B I , LDL - R and LCAT in liver. Results : Compared with the blank group, the serum TC and LDL - C levels were increased significantly while HDL - C level as well as expressions of LDL - R, SR - B I and LCAT in liver were decreased significantly in the model group. In Jianpi Jiangzhi Recipe group the serum contents of TC and LDL - C were lower and HDL - C was higher compared to those of the model group. At the same time,the expressions of LDL -R, SR -B I and LCAT in liver were up -regulated. Conclusion :Jianpi Jiangzhi Recipe could regulate blood lipids, which might be related to the expression of genes associated with cholesterol metabolism.
