摘 要:目的研究参芪十一味颗粒对胃癌放化疗大鼠的免疫功能及不良反应的影响。方法以胃癌Wistar大鼠为模型,并将其随机分为4组,即参芪十一味颗粒组(治疗组)、单纯化疗组(化疗组)、模型组及正常大鼠组(空白组),4组大鼠都给予相应治疗措施且进行观察和数据统计分析。比较评估大鼠的生存质量;采用流式细胞仪检测大鼠静脉血Th1、Th2及其比值;ELISA(enzyme linked immunosorbent assay)法检测IFN-γ和IL-4的含量;测定白细胞(WBC)、血小板(PLT)、血红蛋白(Hb)指标。结果参芪十一味颗粒治疗组大鼠进食量、体质量和生存期较化疗组和模型组明显增加(P<0.05),但仍低于空白组;外周血Th1、Th2及其比值Th1/Th2较化疗组及模型组大鼠均有显著升高(P<0.05);与化疗组和模型组相比,治疗组IFN-γ的含量显著增加,而IL-4含量显著降低(P<0.05);治疗组外周血白细胞、血红蛋白以及血小板减少均显著低于化疗组和模型组(P<0.05)。结论参芪十一味颗粒能有效提高胃癌放化疗大鼠的机体免疫功能,降低毒副作用,改善生活质量,是胃癌放化疗治疗中有效的辅助用药,具有重要的临床使用价值。Objective To study the effect of Shenqi Shiyiwei grain on the immune function and side effects in the chemotherapy-treated rats with gastric cancer. Methods Wistar rats with gastric cancer were selected and randomly divide into 4 groups, namely, as Shenqi Shiyiwei grain group (treatment group ), Chemoradiation group (control group ), the model group and normal rats group.The rats in these 4 groups were given the appropriate treatment measures.The statistical analysis was performed due to the clinical observations and data. The rats' quality of life, including food intake and body weight was evaluated. Thl, Th2, and their ratio in peripheral blood were analyzed by flow cytometry. ELISA (enzyme linked immunosorbent assay) was used for the determination of IFN-γ and IL-4 content.The white blood cells (WBC), platelet (PLT) and hemoglobin (Hb) were determined.Results Shenqi Shiyiwei grain could significantly improve the food intake, body weight and life span of rats in the treated group, compared with those of the control group and the model group (P〈0.05) ; the treatment with Shenqi Shiyiwei grain increased significantly the Thl ,Th2 ,and the ratio of Thl/Tb2 in peripheral blood, compared with those of the chemotherapy group and the model group (P 〈0.05 ). Furthermore, the content of the IFN-γ, in rats of the treatment group was significantly increased, whereas IL-d was significantly decreased ( P 〈0.05) .In addition, the reduction of white blood cells, hemoglobin, and thrombocytopenia in rats of the treatment group peripheral were significant lower than those of the control group and the model group (P〈0.05).Conclusion Shenqi Shiyiwei grain could improve the immune function and the quality of life in chemotherapy-treated rats with gastric cancer and reduce chemotherapy-induced side effects. It is an adjuvant drug for the chemotherapy treatment of rats with gastric cancer and has an important clinical value for using.
