摘  要:为进一步研究绵羊肺腺瘤病毒(JSRV)囊膜蛋白(Env)及其亚基诱导NIH 3T3细胞转化的分子机制,本研究利用JSRV Env及其亚基真核细胞表达质粒分别诱导转化NIH 3T3细胞和稳定表达JSRV受体绵羊Hyal-2的NIH 3T3-Hyal2-HA细胞,并检测转化后细胞的接触抑制特性、Akt的激活和肿瘤相关基因的突变。结果显示,在NIH 3T3细胞中,Env及其穿膜蛋白亚基(TM)均具有诱导细胞转化、激活Akt的能力;而在NIH 3T3-Hyal2-HA细胞中,仅TM具有这样的能力。在细胞转化的过程中,肿瘤相关基因kras、nras、egfr、p53和pik3ca均未发生突变。结果表明,在JSRV Env诱导NIH 3T3细胞转化并激活Akt的过程中,TM亚基可能起主要作用,并且还具有单独诱导NIH 3T3细胞转化的能力;绵羊Hyal-2具有抑制JSRV Env诱导NIH 3T3细胞转化的作用。JSRV Env及其亚基诱导NIH 3T3细胞转化的分子机制可能与肿瘤相关基因突变无关。The aim of this study is to define the molecular mechanisms of transformation induced by jaagsiekte retrovirus (JSRV) envelope protein (Env) and its subunits in NIH 3T3 cells. In this study, eukaryotic expression plasmid of JSRV envelope protein and its subunits were used respectively to induce transformation of NIH 3T3 cells and NIH 3T3-Hyal2-HA cells, besides, contact inhibition characteristics, activation of Akt and mutations in tumor-related genes were detected after the transformation induced. Env and TM could induce transformation and activate Akt in NIH 3T3 cells, however, only TM has these abilities in NIH 3T3-Hyal2-HA cells. The mutations of tumor-related genes, kras, nras, egfr, p53 and pik3ca, did not find. The results showed TM could induce transformation, and it played a major role in JSRV Env induced transformation and Akt activation in NIH 3T3 cells. The ability of JSRV Env in transformation was inhibited by ovine Hayl-2 in NIH 3T3 cells. The mutations of tumor-related genes may not involved in JSRV Env and its subunit induced transformation.
