摘 要:目的 研究Cdc42在纳米化种植体促进成骨细胞功能中的作用.方法 将纳米化种植体与成骨细胞联合培养,通过Western Blot检测Cdc42活性蛋白含量,以及荧光染色检测成骨细胞内F-actin分布,扫描电镜观察成骨细胞形态,MTT检测成骨细胞增殖.结果 纳米化种植体使成骨细胞内Cdc42活性蛋白含量明显增多,Toxin B抑制后其活性明显下降,纳米化种植体表面成骨细胞形态不规则,细胞骨架明显,伸出较多伪足,采用Toxin B处理后细胞形态变圆,伪足减少.成骨细胞增殖功能随种植体纳米化而明显增高,在Toxin B处理后下降.结论 种植体粗化使Cdc42表达增加,可能通过Cdc42调节成骨细胞的黏附及增殖.Objective To investigate the role of Cdc42 in osteoblast function on nanophase-implant.Methods The osteoblast-like cell line MG63 was cultured on normal/nano-phase implants.The levels of activated Cdc42 were measured by the GTPase activity pull-down assay and western blot.Changes of actin cytoskeleton and morphological characteristics were observed by confocal laser scanning microscope (CLSM) and scanning electron microscope (SEM) respectively.Cell proliferation was detected by MTT.Results The activation level of Cdc42 was increase when cultured on nano-phase implant and blocked by Toxin B treatment.The shape of osteoblast attached on implant was irregular with extended pseudopodium.After treated with Toxin B,the shape of cell tended to be round and pseudopodium decreased.The proliferation of osteoblast was increased when cultured on nano-phase implant but decreased after pretreatment by Toxin B.Conclusion Nano-phase implant could increase the impression of active Cdc42.Nano-phase implant might improve the proliferation and inhesion of osteoblast through Cdc42.
