摘 要:目的:本实验以载脂蛋白基因缺陷(ApoE-/-)小鼠为研究对象,观察其在动脉粥样硬化(AS)发病过程中有氧运动对脂联素和C反应蛋白(CRP)的影响,以期探讨有氧运动通过脂联素-CRP发挥抗AS作用的机制。方法:8周龄雄性ApoE-/-小鼠20只,随机分为安静组(C)10只和运动组(E)10只。两组小鼠均饲以高脂饲料,C组不训练,E组小鼠进行14周跑台运动。14周末取材比较两组小鼠主动脉管壁及斑块的病理变化,检测比较其脂肪组织脂联素mRNA表达、血清脂联素水平及主动脉CRP表达、血清CRP水平。结果:E组小鼠与C组小鼠相比,主动脉血管壁纤维弹力板和内膜较完整,AS斑块面积相对较小。E组小鼠脂肪组织脂联素mRNA表达及血清水平较C组显著增加,主动脉CRP的表达及血清水平较C组小鼠则有显著降低,且主动脉CRP的表达与脂联素mRNA表达及血清水平呈显著负相关。结论:有氧运动可以通过提高血清脂联素、脂联素mRNA表达水平的途径,有效降低血清CRP和主动脉CRP的表达,从而发挥延缓AS发展的作用,这可能是运动通过脂联素-CRP发挥抗AS作用的另一机制。Objective: To approach the effects and meclaanlsm oI aerot)lC exercise on atucrosclerosis(AS) and the effect of adiponectin-C-reactive protein (CRP) in apolipoprotein E(ApoE)-deficient mice AS model. Method: Eight-week-old ApoE-deficient mice were randomly assigned to two groups: ApoE-deficient mice control group (C); aerobic exercise-trained group (E). The mice were fed by high fat diet. Mice in aerobic exercise-trained groups ran on a treadmill for a total of 14 weeks, At the end of experiment, AS was observed. Adiponectin rnRNA in adipose tissue and serum adiponectin were tested. CRP in aortic and serum CRP were detected. Result: The AS of E group was significantly better than that of C group; Adiponectin-mRNA in adipose tissue and serum adiponectin of E group were significantly higher than that of C group. The CRP and serum CRP were significantly down-regulated in E group. The CRP and adiponectin-mRNA were significant negative correlated. Conclusion: It suggested that 14 weeks aerobic exercise could significantly increase adiponectin-mRNA in adipose tissue and significantly decreased CRP in aortae. Aerobic exercise was likely to one of the factor by the adiponectin-CRP to anti-atherogenesis. Author's address Physical Education College of Zhengzhou University, 450044.
