贝丹益肺胶囊对肺纤维化大鼠肺组织TGFβ-Smads表达的影响

摘　　要：目的:观察贝丹益肺胶囊对肺纤维化大鼠肺组织转化生长因子β(TGF-β1)信号通路分子mRNA表达的影响,探讨其防治肺纤维化的作用及机制。方法:气管内滴入博来霉素(5mg/kg)建立大鼠肺纤维化模型。将40只Wistar大鼠随机分为四组(每组10只):对照组、肺纤维化模型组、强的松组和贝丹益肺胶囊组。HE染色检测肺组织病理改变,原位杂交技术检测肺组织Smad4 mRNA表达,实时荧光定量RT-PCR技术检测TGF-β1mRNA表达。结果:胶原纤维染色显示模型组大鼠肺内胶原纤维表达显著高于对照组和贝丹益肺胶囊治疗组(P<0.05)。大鼠肺内TGF-β1及Smad4 mRNA表达在模型组显著高于对照组(P<0.01),在贝丹益肺胶囊治疗组显著低于模型组(P<0.05)。结论:贝丹益肺胶囊能有效地减轻肺纤维化大鼠肺组织的纤维化程度,其作用机制主要是通过抑制TGF-β1及Smad4mRNA表达上调,从而干扰TGF-β1/Smad4信号通路对靶基因的激活来实现的。Objective：To observe Beidan Yifei capsule on lung tissues of rats with pulmonary fibrosis transforming growth factor β（TGF -β） influencing the expression of mRNA signaling molecules ,and to discuss its function and mechanism in treating pulmonary fibrosis.Method：Intratracheal instillation of bleomycin （5 mg/kg）was to establish the rat models with pulmonary fibrosis.40 Wistar rats were randomly divided into four groups （n =10 in each group）： control group, pulmonary fibrosis model group, prednisone group and Beidan Yifei capsule group .HE staining of lung pathology was detected; detect Smad4 expression of mRNA in lung tissues in situ hybridization ; real -time fluorescence quantitative RT -PCR was used for the detection of expression of TGF -β1mRNA.Results：Collagen fiber staining showed that the expression of collagen fibers in rats of model group was significantly higher than that in the control group and Beidan Yifei capsule group （P ＆lt;0.05）.TGF -β1 and Smad4 mRNA expression in lungs of rats in the model group were sig -nificantly higher than those in the control group （P ＆lt;0.01）,and they were significantly lower in Beidan Yifei capsule group than those in the model group （P ＆lt;0.05）.Conclusion：Beidan Yifei capsule can effectively alleviate the lung tis -sue of rats with pulmonary fibrosis.Its mechanism is mainly through the inhibition of TGF -β1 and Smad4 expression of mRNA, which interferes with TGF -β1 /Smad4 signal pathway activation of target genes .
