摘  要:目的:探讨慢性束缚应激对大鼠在旷场迷宫中的行为学参数和大脑杏仁核部位5-羟色胺(5-HT)、5-羟吲哚乙酸(5-HIAA)含量的影响,以及加味甘麦大枣汤的干预作用。方法:采用经典的慢性束缚应激的实验方法,复制大鼠慢性应激的模型,然后记录大鼠在旷场迷宫中的行为学参数;分离大鼠大脑杏仁核部位,采用酶联免疫吸附法测定其中5-HT、5-HIAA的浓度,进而推算出慢性束缚应激对杏仁核部位5-HIAA/5-HT比值的影响,并探究加味甘麦大枣汤对其的干预作用。结果:实验结果表明,与空白组相比,模型组大鼠在穿格数、直立次数和理毛次数这三个行为学参数上明显减少(P<0.05),而排便次数这个参数则显著增加(P<0.05);杏仁核部位5-HT、5-HIAA含量以及5-HIAA/5-HT比值明显减小(P<0.05)。与模型组相比,给药组大鼠在穿格数和理毛次数这两个指标上显著增加(P<0.05),排便次数明显减少(P<0.05);在直立次数这个指标上虽有增加的趋势,但无统计学意义(P>0.05)。杏仁核部位5-HT、5-HIAA的含量显著升高(P<0.05),虽然5-HIAA/5-HT的比值有升高的趋势,但是无统计学上的差异(P>0.05)。结论:慢性束缚应激使大鼠在旷场迷宫中的各行为学参数明显改变,使大鼠杏仁核部位5-HT和其代谢产物的含量以及5-HT能神经系统的兴奋性降低。中药复方加味甘麦大枣汤能明显对抗慢性束缚应激对大鼠造成的影响。Objective:To investigate the effect of chronic restraint stress on rats ’ behavioral parameters in open field maze and the levels of 5 -HT, 5 -HIAA in amygdala, and the intervention of Jiawei Ganmai Dazao Tang (JWGMDZT). Methods: In this research, chronic restraint stress method was used to establish the model of chronic mild stress in rats . Behavioral parameters of rats in open field maze and the levels of 5 -HT, 5 -HIAA, 5 -HIAA/5 -HT in amygdala were evaluaed.The intervention of JWGMDZT was also investigated .Results: Compared with vehicle, the number of crossing line, rearing and grooming in open field maze significantly reduced (P &lt;0.05), while defaecation increased markedly(P &lt;0.05).The levels of 5 -HT, 5 -HIAA and 5 -HIAA/5 -HT in amygdala reduced significantly (P &lt;0.05).Compared with model group, the number of crossing line and grooming significantly increased (P &lt;0.05), while defaecation reduced markedly(P &lt;0.05).Though there was an increased trend of the number of rearing , no signifi-cance was performed(P &gt;0.05).The levels of 5 -HT and 5 -HIAA in amygdala increased significantly (P &lt;0.05). Though there was an increased trend of 5 -HIAA/5 -HT, no significance was detected by ANOVA(P &gt;0.05).Con-clusion: The behavior and excitability of 5 -HT energetic nervous system in the amygdala of rats were changed by chron -ic restraint stress.JWGMDZT could effectively alleviate the impact of chronic restraint stress on rats .
