摘 要:目的观察中药复方清热颗粒对多重耐药阴沟肠杆菌(Ecl)主动外排泵机制的影响。方法 (1)含药血清的制备:选购健康大耳白兔,随机分为两组,试验组6只和空白对照组6只,适应性喂养1周。实验组予以无菌复方清热颗粒剂药液灌胃,每日2次,连续给药5 d;对照组予同等体积灭菌蒸馏水灌胃相同时间及给药次数。末次给药后60 min,无菌心脏采血,分离含药血清和空白血清,于-20℃保存备用。(2)应用外排泵抑制剂碳酰氰基-对-氯苯腙(CCCP)筛选出外排泵表型阳性多重耐药Ecl 10株,利用琼脂对倍稀释法测出环丙沙星,对上述Ecl的最小抑菌浓度(MIC)值,并用同样方法测出含药血清、空白血清、复方清热颗粒、CCCP及分别联合环丙沙星对上述10株Ecl的最小抑菌浓度(MIC)值变化。结果复方中药清热颗粒含药血清可降低环丙沙星对多重耐药EclMIC值。结论中药复方清热颗可抑制多重耐药Ecl的耐药情况。Objective: To explore the effect of Compound Qingre Granules on efflux pumps of multidrug-resistant enterobaeter cloacae. Methods: Ⅰ. The preparation of drug serum: healthy rabbits with big ears were selected and randomly divided into two groups : 6 in experimental group and 6 in control group. After one week of adaptive feeding,experimental group was given Compound Qingre Granules by intragastric administration twice a day for 5 days. The control group was given sterile purified water in the same way with the same dose. Blood was drawn from heart after 60 minutes at the last medication and was centrifuged to separate drug serum and blank serum ,which were then conserved in -20 ℃. Ⅱ. 10 strains of efflux pump positive multidrug-resistant enterobacter cloacae were selected by using CCCP. The minimal inhibitory concentration of ciprofloxacin was measured within and withiout Compound Qingre Granules,drug serum,blank serum and CCCP by agar dilution method. Results: Compound Qingre Granules could significantly reduce the MIC of ciprofloxacin to multidrug-resistant enterobaeter cloacae. Conclusions: Compound Qingre Granules can alleviate the resisitance of muhidrug-resistant enterobacter cloacae.
