加味茯苓泽泻汤对高脂血症大鼠血清NO和MPO水平的影响  

Effects of Jiawei Fuling Zexie Decoction on Levels of Seral Nitric Oxide and Myeloperoxidase in Hyperlipidemia Rats

摘　　要：研究加味茯苓泽泻汤对高脂血症(HLP)SD大鼠血清一氧化氮(NO)含量和髓过氧化物酶(MPO)活性的影响,并探讨其治疗高脂血症的作用机制。健康SPF级雄性SD大鼠60只,随机选取10只作为空白组,其余作为高脂饮食组;高脂饮食组采用腹腔注射维生素D3和经口灌胃高脂乳剂复制高脂血症大鼠模型,造模成功后随机选取40只平分为模型组、加味茯苓泽泻汤高、低剂量组和辛伐他汀组,每组各10只,后3组分别给予相应的药物治疗;28d后,检测各组大鼠治疗前后的血脂指标、血清NO含量和MPO活性水平。与模型组比较,加味茯苓泽泻汤高、低剂量组的血清胆固醇(TC)、甘油三脂(TG)、低密度脂蛋白-胆固醇(LDL-C)及血清MPO活性水平均显著降低(P<0.05),而高密度脂蛋白-胆固醇(HDL-C)及血清NO含量水平均显著升高(P<0.05)。加味茯苓泽泻汤高剂量组对大鼠的血脂指标、NO含量和MPO活性水平的影响效果基本与辛伐他汀组等同。试验结果提示,加味茯苓泽泻汤通过抑制MPO的活性,升高NO的含量,进而对血管内皮功能的完整性起到一定的保护作用。To investigate the effect of Jiawei Fuling Zexie Decoction（JWFLZXD）on the levels of serum ni-tric oxide content and myeloperoxidase activity in hyperlipidemia rats,and study its therapeutic mechanism in the treatment of hyperlipidemia,60 health SPF male SD rats were used,10 rats were randomly selected as the blank group,and the rest as a high-fat diet group.After the modeling successfully with vitamin D3 and high-fat emulsions,randomly selected 40 rats in high-fat diet group were equally divided into model group,JWFLZXD high-dosage group,low-dosage group and the simvastatin group,every groups contain 10 rats.The rats in the three experiment groups were given appropriate medication for 28 days.The seral lipid parameters,MPO and NO were detected in rats before and after treatment.Compared with the model group,the levels of TC,TG,LDGL-C and MPO in the JWFLZXD high-dosage group and low-dosage group were significantly reduced（P 〈0.05）.The levels of HDL-C and NO were significantly raised（P 〈0.05）.The effects of JWFLZXD high-dosage group on the lipid parameters,the content of NO,the active of MPO and the liver index in rats were almost equivalent to the simvastatin group＇s.The result revealed that JWFLZXD can inhibit the activitity of MPO to increase NO levels,and thus to play a protective role on the integrity of the vascular endothelial function.
