摘 要:目的研究熊胆粉在麝香通心滴丸抗动脉粥样硬化中的作用,为其凉开法治疗特点及临床推广应用提供实验数据。方法采用特异性基因敲除小鼠结合高脂饮食复制动脉粥样硬化动物模型,灌服熊胆粉水溶液,并以麝香通心滴丸全方水溶液及全方去熊胆粉水溶液为对照,通过HE染色、油红O染色及Masson’s Trichrome染色,观察对主动脉及主动脉根动脉粥样硬化斑块形成的干预作用。生化法测定血清中总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白(HDL)及低密度脂蛋白(LDL)水平。结果与正常组对比,模型组斑块损伤面积显著增加(P<0.01),血清TG、TC和LDL均显著升高(P<0.01),HDL明显下降(P<0.01);与模型组对比,各给药组斑块损伤面积均显著减小(P<0.05),全方组作用优于去熊胆粉组。各给药组与模型组对比,血清TG、TC均显著下降(P<0.01),HDL明显上升(P<0.01),全方组作用优于去熊胆粉组,此外,熊胆粉组降低LDL作用显著(P<0.01)。结论熊胆粉在麝香通心滴丸抗动脉粥样硬化作用中具有突出的贡献,特别是在降低LDL作用中贡献显著。Objective To study the anti atherosclerotic activity of bear bile powder (BBP)in Shexiang Tongxin dripping pills (STDP)on ApoE / mice.Methods ApoE / mice fed with high fat diet were utilized to reproduce animal models of atheroscle-rosis.ApoE / mice were treated with bear bile powder aqueous solution,STDP solution and STDP depleted with bear bile power so-lution,respectively.Interventional effect on the atherosclerotic lesion size of aorta and aortic root were measured by hematoxylin&eosin staining,oil red O staining and masson’s trichrome staining,respectively.Serum levels of total cholesterol (CHO),triglyceride (TG), high density lipoprotein (HDL)and low density lipoprotein (LDL)were determined by biochemical method.Results Compared to that from the ApoE / mice treated with vehicle,three treated groups were marked with a significant decrease in the size and extent of atherosclerotic lesions(P<0.05).As expected,a better therapeutic effect was observed in STDP treated mice compared to that from the mice receiving STDP depleted with bear bile power.Compared to that from the ApoE / mice treated with vehicle,in the serum levels of CHO and TG levels was decreased(P<0.01),increased serum HDL (P<0.01)was also noted in three groups of mice receiving either BBP,STDP or STDP without BBP.Furthermore,lower level of LDL was noted in BBP treated mice.Conclusion A BBP contributes significantly in mediating the anti atherosclerotic effects of STDP,in particular,in reducing serum LDL level in ApoE / mouse model.
