摘 要:目的:本研究旨在探讨UC患者外周血单个核细胞NF-κB及磷酸化NF-κB及相关细胞因子在溃疡性结肠炎(UC)中的表达,并进一步探讨体外黄芩苷干预的作用。方法:应用western blotting蛋白水平检测UC患者外周血单个核细胞NF-κB p65和Phospho-NF-k B p65,用ELISA法检测血清IFN-r、IL-4、IL-6、IL-10水平及体外黄芩苷干预下的检测上述指标。结果:与健康对照组和IBS-D组相比较,p-NK-FB明显增高,NK-FB明显下降,p-NK-FB/NK-FB明显增高,差异有显著性(P<0.05);IBS-D组和健康对照组之间无显著性差异(P>0.05)。体外培养体外黄芩苷干预下,在40umol浓度干预下p-NK-FB/NK-FB较未干预前、DMSO干预、5umol干预、10umol和20umol浓度干预下下降,存在显著性差异(P<0.05)。从ELISA检测较高浓度的黄芩苷能够明显抑制IFN-r、IL-6等细胞因子,可以使IL-4、IL-10升高。结论:黄芩苷作为黄芩汤的重要组成成分之一,可能是一种潜在的免疫抑制剂,可能抑制p-NK-FB/NK-FB,调节免疫平衡而缓解溃疡性结肠炎的炎症反应,为深入研究UC的发病机制及新药开发提供理论基础。Objective: The purpose of this study is to investigate the UC patients peripheral blood mononuclear cells NF-kappaB andphosphorylation of NF-kappaB and expression of related cytokines in ulcerative colitis (UC), and to further explore the in vitro baicalininterventioneffect. Methods: Application Western blotting protein levels in detection of patients with UC peripheral blood mononuclearcells NF-κB p65 and phospho-NF-kB p65 detected by ELISA in serum IFN-R, IL-4, IL-6, IL-10 level and in vitro baicalin interfere withthe detection of these indicators. Results: Compared with the healthy control group and the IBS-D group compared with p-NK-FBsignificantly increased the NK-FB decreased p-NK-FB/NK-FB the significantly higher, the difference was significant (P〈0.05);nosignificant difference (P〉0.05) between the IBS-D group and the healthy control group. Cultured in vitro in vitro baicalin in theconcentration of 40umol intervention under p-NK-FB/NK-FB than non-intervention, a DMSO intervention, 5umol intervention, 10umoland 20umol concentration intervention decline, there is a significant difference (P〈0.05) . ELISA detected a higher concentration ofbaicalin notably the inhibition of IFN-r, IL-6 and other cytokines, can make IL-4, IL-10 increased. Conclusion: baicalin as one of theimportant components of HQT, may be a potential immunosuppressive agents, may inhibit p-NK-FB/NK-FB regulation of the immunebalance and relieve ulcerative colon inflammation reaction, for in-depth UC pathogenesis and development of new drugs to provide atheoretical basis.
