摘 要:目的:探讨抵当汤对高脂饮食胰岛素抵抗(IR)大鼠血栓素A2(TXA2)、6-酮-前列腺素(6-Keto-PGF1a)的影响。方法:大鼠35只随机分为5组,空白对照组每日给予普通饲料和蒸馏水(10ml/kg)灌胃;模型组每日给予高脂饲料和蒸馏水(10ml/kg)灌胃;抵当汤低剂量组每日给予高脂饲料和抵当汤药液(1g/kg)灌胃;抵当汤高剂量组每日给予高脂饲料和抵当汤药液(2g/kg)灌胃;血脂康组每日给予高脂饲料和血脂康(0.2g/kg)灌胃,连续喂养8周。取血浆、血清标本,检测大鼠空腹血糖、空腹血清胰岛素、胰岛素抵抗指数、胰岛素敏感指数、血栓素、6-酮-前列环素。结果:模型组与空白组相比,大鼠血清Glu、FINS、HOMA-IR水平明显升高,ISI显著降低,TXB2含量显著升高,6-Keto-PGF1a含量显著下降;抵当汤组(1g/kg)、抵当汤组(2g/kg)与模型组相比,大鼠HOMAIR均明显降低,而ISI明显升高;TXB2含量显著降低;抵当汤组(1g/kg)。6-Keto-PGF1a含量显著升高。结论:大鼠IR模型可通过喂饲高脂饲料成功建立,表明IR与血脂异常存在着密切的关系,抵当汤能改善IR大鼠胰岛素敏感性,表明抵当汤可防治IR,调节血脂,防治IR大鼠脂代谢紊乱。抵当汤可维持IR大鼠TXA2和6-Keto-PGF1A的平衡。Objective: To observe the effects of Didangtang on TXA2and 6-Keto-PGF1ain rat model with insulin-resistant induced by high fat die.Methods: 35 health and clean level Wistar male rats were chosen and randomly divided into 5 groups,control group,model group,low-dose Didangtang group,high-dose Didangtang group and Xuezhikang group with 7 rats each. The rats of control group were fed with normal diet and distilled water( 10ml /kg) daily,model group was fed high fat diet and distilled water( 10ml /kg) daily,low-dose Didangtang group was fed high fat diet and Didangtang( 1g /kg) daily,high-dose Didangtang group was fed high fat diet and Didangtang( 2g /kg) daily,Xuezhikang group was fed high fat diet and Xuezhikang( 0. 2g /kg) daily. After 8 weeks of feeding,take blood by decapitate. Blood was taken to measure biochemical indexs,TXA2and 6-Keto-PGF1a. Results: Didangtang can markedly decrease TXA2and increase 6-Keto-PGF1ain Insulin Resistance Rats. Conclusion: Didangtang can keep the balance of TXA2and 6-Keto-PGF1ain IR rats. Didangtang is an ideal medicine in preventing and treating dyslipidemia.
