摘  要:目的:探讨高车前苷对棕榈酸诱导的人脐静脉内皮细胞凋亡的保护作用及其机制。方法:建立棕榈酸诱导的人脐静脉内皮细胞凋亡模型,荧光显微镜下观察细胞凋亡形态,以Annexin V-FITC染色、流式细胞仪检测细胞凋亡率,并用荧光显微镜检测细胞内活性氧和一氧化氮的水平。结果:高车前苷(1,10μmol/L)明显抑制由棕榈酸诱导的内皮细胞凋亡,降低内皮细胞内活性氧的水平和增加细胞内一氧化氮的生成。结论:高车前苷对棕榈酸诱导的内皮细胞凋亡具有保护作用,其作用机制与抗氧化应激有关。Objective: To study the protective effect and its mechanism of homoplantagnin on human umbilical vein endothelial cells apoptosis induced by palmitic acid( PA). Methods: HUVECs apoptosis model was established by PA. Morphology of apoptotic cells was detected by fluorescence microscope and the apoptosis rate was analyzed by flow cytometry. Fluorescence recording of intracellular fluorescence intensity of reactive oxygen species and nitric oxide were tested. Result: Homoplantagin( 1,10 μmol /L) significantly inhibited the endothelial cells apoptosis induced by PA,reduced the intracellular ROS and increased NO generation. Conclusion: These results suggest that homoplantaginin can alleviate endothelial cells apoptosis induced by PA which is probably due to its anti-peroxidative effect.
