摘 要:目的:研究山茱萸效应成分马钱苷、莫诺苷对晚期糖基化终末产物(AGEs)损伤人脐静脉内皮细胞(HUVEC)增殖的影响及保护作用。方法:体外培养HUVEC,加入阳性药氨基胍及马钱苷、莫诺苷(终浓度分别为0.05、0.1、0.5、1、5、10μmol/L),预孵1h后,加入终浓度为200 mg/L的AGEs,另设模型组及空白对照组,孵育24h后,采用MTT法检测吸光度(OD)值;体外培养HUVEC,加入氨基胍及马钱苷、莫诺苷(终浓度分别为1、10、100μmol/L)预孵1h后,加入终浓度为200 mg/L的AGEs,另设模型组及空白对照组,孵育24h后取细胞培养上清液,检测活性氧(ROS)、超氧化物歧化酶(SOD)、丙二醛(MDA)、一氧化氮(NO)、内皮素(ET)、血管紧张素Ⅱ(AngⅡ)、转化生长因子-β(TGF-β)、白介素-1β(IL-1β)水平。结果:对增殖影响研究发现,氨基胍在终浓度5、10μmol/L时,马钱苷在终浓度1、5、10μmol/L时,莫诺苷在终浓度0.1、0.5、1、5、10μmol/L时,对AGEs损伤HUVEC具有显著的增殖保护作用,且随着浓度增高,增殖保护作用增强;对保护作用研究发现,模型组细胞上清液中SOD、NO含量降低,ROS、MDA、ET、AngⅡ、TGF-β、IL-1β含量增加;而马钱苷、莫诺苷可不同程度的提高SOD、NO活力,降低ROS、MDA、ET、AngⅡ、TGF-β、IL-1β活力,与模型组比较有显著性差异。结论:山茱萸效应成分马钱苷、莫诺苷可通过调节内皮细胞分泌功能、抑制氧化应激、炎症因子释放及抗细胞粘连等保护血管内皮细胞最终达到治疗糖尿病目的。Objective: To investigate the effects of loganin and morronside from effective ingredients of Cornus officinalis protect HUVEC damaged by AGEs. Methods: HUVEC was cultured in vitro and administrated by aminoguanidine( treated as positive drug),loganin and morronside(final concentrations are 0. 05,0. 1,0. 5,1,5,10μmol /L). After incubating 1h,added in AGEs which the final concentration is 200 mg /L,At the same time set the model group and normal group. After incubating 24h,the OD values were measured by using MTT method. HUVEC was cultured in vitro and administrated by aminoguanidine,loganin and morronside( final concentrations are 1,10,100μmol /L). After incubating 1h,added in AGEs which the final concentration is 200 mg /L,At the same time set the model and normal groups. After incubating24h,the supernatant was extracted and the levels of ROS,SOD,MDA,NO,ET,AngⅡ,TGF-β,IL-1β were measured by the corresponding kits. Results: For the effects of proliferation,we found aminoguanidine( final concentrations are 5,10μmol /L),loganin( final concentrations are 1,5,10μmol /L) and morronside( final concentrations are 0. 1,0. 5,1,5,10μmol /L) could protect HUVEC damaged by AGEs; For the effects of protection,we found the contents of SOD and NO from model group decreased( P 〈 0. 05,P 〈 0. 01) and the contents of ROS,MDA,ET,AngⅡ,TGF-β,IL-1β increased( P 〈 0. 05,P 〈 0. 01); however,loganin and morronside could increase the SOD and NO contents and reduce the ROS,MDA,ET,AngⅡ,TGF-β,IL-1β contents in different extents. Conclusion: loganin and morronside could protect vascular endothelial cells by improving endothelial cell function,inhibiting oxidative stress,resisting inflammatory cytokines and cell adhesion,and at last improving diabetes mellitus.
