吴茱萸碱改善CCl_4引起的小鼠肝纤维化损伤研究

摘　　要：目的:研究吴茱萸碱的抗肝纤维化作用。方法:雄性C57鼠采用腹腔注射CCl4构建肝纤维化模型,同时采用吴茱萸碱(18mg/kg、9mg/kg)、扶正化瘀胶囊(750mg/kg)进行灌胃给药,干预运HE染色检测肝纤维化的变化;生化检测血清丙氨酸转移酶(ALT)和天冬氨酸转移酶(AST),以及肝组织的SOD、MDA含量;Western blot法检测转化生长因子β(TGF-β)及血小板衍生生长因子(PDGF)的蛋白表达。结果:吴茱萸碱(18mg/kg、9mg/kg)组和扶正化瘀胶囊(750mg/kg)组显著抑制CCl4诱导的小鼠肝纤维化程度,血清ALT、AST水平及肝脏组织中MDA水平明显降低,SOD显著提高;吴茱萸碱(18mg/kg、9mg/kg)组和扶正化瘀胶囊(750mg/kg)组显著抑制肝组织中TGF-1和PDGF的蛋白表达。结论:吴茱萸碱具有显著的改善肝纤维化作用。Objective： To investigate the anti-hepatofibrotic effects of evodiamine on liver fibrosis. Methods： Male C57mice were induced with20% CCl4（ 2ml/kg） for 6weeks to induce liver fibrosis,and were treated with intragastrical gavage of 18and 9mg/kg evodiamine for14d since the fifth week of CCl4induction. Liver fibrosis of the mice was evaluated by HE stain. Activities of serum alanine aminotransferase（ ALT）and aspartate aminotransferase（ AST）,and liver content of SOD and MDA were measured with automated biochemistry analyzer. The protein levels of transforming growth factor beta（ TGF-β） and PDGF were detected by Western blot analysis. Results： Evodiamine（ 18mg /kg,9mg /kg） groups significantly attenuates the progression of liver fibrosis induced by CCl4. Treatment with evodiamine（ 18mg /kg,9mg /kg） groups also resulted in significant decreases in the serum levels of ALT and AST,as well as the content of MDA and increased SOD. Evodiamine（ 18mg/kg,9mg/kg） groups significantly reduced the protein expression levels of TGF-βand PDGF in the liver. Evodiamine exerts anti-fibrotic effects on the liver induced by CCl4.
