摘  要:目的:观察荞麦黄酮对脑缺血再灌注大鼠损伤的影响及其可能机制。方法:SD大鼠随机分为假手术组、模型对照组、荞麦黄酮剂量组(100、200、400mg/kg)及尼莫地平组(6mg/kg),各组灌胃给药1次/d,连续5d,末次给药2 h后,建立大鼠大脑中动脉闭塞再灌注模型,再灌注24 h后将各组大鼠进行神经行为评分,并测定大鼠脑梗死面积、脑含水量;测定血清中超氧化物歧化酶(SOD)、丙二醛(MDA)、白细胞介素-1β(IL-1β)、白细胞介素-10(IL-10)及肿瘤坏死因子-α(TNF-α)、内皮素(ET)、诱生型一氧化氮合酶(iNOS)及一氧化氮(NO)含量。结果:与假手术组比较,模型对照组大鼠神经行为评分值、脑梗死面积、脑含水量、MDA、IL-1β、TNF-α、ET、iNOS及NO含量均显著升高,而SOD及IL-10含量下降。与模型对照组比较,荞麦黄酮各剂量组(100、200、400mg/kg)能不同程度改善上述指标。结论:荞麦黄酮在100~400 mg/kg剂量范围内对脑缺血再灌注损伤大鼠具有一定保护作用,其机制可能与抗氧化、抗炎及降低血管活性物质ET、NO含量相关。Objective: To investigate protective effect and mechanism of buckwheat flavones( BF) cerebral ischemia reperfusion injury in rats and explore possible mechanisms. Methods: SD rats were randomly divided into Sham group,model group,BF groups with various dosages( 100,200 and 400 mg /kg) and nimodipine group( 6 mg /kg). The rats were given drugs by ig administration once per day,for 5 d. At 2 h after last administration,the model of cerebral ischemia in rats was made by middle cerebral artery occlusion-reperfusion. At 24h after perfusion,the behavior,cerebral infarct size and water content of cerebrum in rats were determined,and SOD,MDA,IL-1β,IL-10,TNF-α,ET,iNOS and NO levels in serum were tested. Results: Compared with sham group,values of nerve behavior,cerebral infarct size,water content of cerebrum,MDA,IL-1β,TNF-α,ET,iNOS and NO significantly increase,while levels of SOD and IL-10 decrease( P 〈 0. 01). Compared with model group,BF with various doses( 100,200,400 mg /kg) could improve above each index( P 〈 0. 05,P 〈 0. 01). Conclusion: BF in the dose of 100 ~ 400 mg /kg has protective effect on cerebral ischemia reperfusion injury in rats,its possible mechanisms are in part associated with antioxygenation and anti-inflammatory action and decrease the levels of vascular active substances such as ET,NO.
