瘤果黑种草子总皂苷治疗类风湿性关节炎的作用研究

摘　　要：目的:考察瘤果黑种草子总皂苷提取物抗炎镇痛的药效作用,为其治疗类风湿性关节炎提供依据。方法:采用二甲苯致小鼠耳肿胀、冰醋酸致小鼠扭体、冰醋酸致小鼠腹腔毛细血管通透性增高模型,Freund's完全佐剂致大鼠原发性、继发性关节炎损害和牛Ⅱ型胶原诱导大鼠胶原性关节炎等模型,对照观察瘤果黑种草子总皂苷提取物的抗炎、镇痛等作用。结果:瘤果黑种草子总皂苷提取物在6~24mg/kg均能能明显减少冰醋酸致小鼠扭体反应次数,均能减轻二甲苯致小鼠耳肿胀,均能抑制小鼠腹腔毛细血管通透性增高,均在24mg/kg剂量的抑制率高于野木瓜片;在4.2~16.6mg/kg对完全佐剂所致大鼠原发性和继发性炎症及Ⅱ型胶原诱导的大鼠胶原性关节炎均有一定抑制作用,佐剂所致大鼠原发性损害瘤果黑种草子总皂苷8.3~16.6mg/kg剂量作用均较野木瓜片组好,病理显示与模型组比较,各给药组炎性症状均有所减轻,但野木瓜片组与瘤果黑种草子总皂苷各剂量组没有显著性差异;佐剂所致大鼠继发性损害16.6mg/kg剂量组则在第25d^29d能明显减轻大鼠左后足肿胀度;胶原性关节炎16.6mg/kg剂量组左后足和右后足肿胀度在致炎后第17~26d均低于野木瓜片组。结论:瘤果黑种草子总皂苷提取物有良好的抗炎镇痛和治疗关节炎的作用。Aim： To study the pharmacodynamics of total sapionins from Nigella glandulifera seeds（ NGS） on anti-inflammatory analgesic,provide the basis for the treatment of arthritis. Methods： The anti-inflammatory and analgesic effects were studied by applying the models of oxyleneinduced swelling of ear in mice,acetic acid-induced twisting reaction in mice,higher capillary permeability was caused by acetic acid,the primary and secondary arthritis of rats induced by the complete Freund’s adjuvant and CIA model in rats was aroused by injection of bovine collagen type Ⅱ（ BCⅡ） emulsion. Results： The total sapionins from NGS in the 6 ~ 24 mg /kgod dose could obviously decrease twisting times of mice caused by acetic acid,alleviate the ear edema induced by oxylene in mice,and nhibit increasing capillary permeability,inhibition rate of all in the 24 mg /kgod dose was higher than Ye Mugua tablet. It in the 4. 2 ~ 16. 6mg /kgod dose could inhibit the primary,secondary and CIA inflammation of rats induced by CFA and BCⅡ,respectively. Its effects in the 8. 3 ~ 16. 6mg /kgod dose were better than Ye Mugua tablet to the primary arthritis of rats induced by the CFA. Pathology displayed inflammatory symptoms of the treatment group were all reduced than the model group,but there was no significant difference in positive control group and test group. It could significantly reduce left hind paw swelling of the rat in 16. 6mg /kgod dose at 25 d ~ 29 d to the secondary arthritis of rats induced by the CFA. The left and right hind paw swelling in 16. 6mg /kgod dose were lower than Ye Mugua tablet at 17 d ~ 26 d to arthritis of rats induced by BCⅡ. Conclusion： The total sapionins from NGS has the anti-inflammatory analgesic and effects.
