七十味珍珠丸抗大鼠脑血栓形成作用及机制研究

摘　　要：目的:观察七十味珍珠丸抗实验性大鼠脑血栓形成的作用,并初步探讨其作用机制。方法:用结扎颈外动脉同时颈内动脉注射复合血栓诱导剂的方法建立实验性脑血栓形成大鼠模型。以阿司匹林作为阳性对照组,观察七十味珍珠丸对实验性脑血栓大鼠脑梗塞区伊文思蓝含量、脑水肿程度、血液流变学指标及血浆中血栓素A2(thromboxane A2,TXA2)、前列环素(prostacyclin,PGI2)水平的影响。结果:与模型组相比,七十味珍珠丸高剂量(300 mg/kg)、中剂量(150 mg/kg)、低剂量(75 mg/kg)都不同程度的减轻了实验性模型大鼠脑血栓程度、脑水肿程度;降低了全血低切粘度、血浆粘度、红细胞聚集指数;同时抑制了模型大鼠血浆中TXA2和PGI2这对活性物质的失衡。结论:七十味珍珠丸可抑制血栓诱导剂诱导的脑血栓形成;其机制可能与下调血液流变学指标和改善TXA2/PGI2系统失衡有关。Objective： To explore the inhibitory effect of Rannasangpei on cerebral thrombosis in rats and its mechanism involved. Methods： The cerebral thrombosis model was performed by ligation of the branch of external jugular arteria and injecting the thrombosis inducer 1ml /kg into the internal carotid arteria. With aspirin enteric-coated tablets as positive controls,the effects of Rannasangpei on the contents of Evans-blue in brain tissue with thrombus,the extents of brain edema,the hemorheology indexes and the changes of TXA2and PGI2contents in rats plasma were observed. Results： Compared with the model group,the extents of the rats cerebral thrombus and brain edema in three Rannasangpei intervention groups were reduced in different degrees（ P 〈 0. 01）,meanwhile,the unbalance of TXA2and PGI2in that groups was significantly inhibited（ P 〈 0. 01）. The hemorheology indexes of Rannasangpei intervention groups which included the low cut blood viscosity,the plasma viscosity and erythrocyte aggregation index were obviously decreased than that of the model group（ P 〈 0. 01 or P 〈 0. 05）. Conclusion：Rannasangpei has significant inhibitory effect on cerebral thrombosis in rats,its protective mechanism may come from effectively lowering the hemorheology indexes and improving the disorder of TXA2/PGI2system.
