摘 要:目的:制备一种具备乳腺癌靶向性的超小超顺磁性氧化铁颗粒(USPIO),与病理组织学相对照验证其活体靶向效果。方法化学共沉淀一步法制备 USPIO,将其与乳腺癌特异表达的人乳腺珠蛋白(hMAM)单抗通过共价键耦联,制备具有乳腺癌靶向性的 USPIO,并测定其粒径、铁含量。比较其与非靶向 USPIO 在荷瘤动物模型中磁共振(MRI)信号强度(SI)差异及肿瘤细胞铁标记率差异。结果抗 hMAM-USPIO 平均粒径(50.37±13.10)nm,铁含量20.18 mg/mL。荷瘤动物模型 MRI:抗 hMAM-US-PIO 组肿瘤较非靶向 USPIO 组 SI 降低更显著(P 〈0.05)。肿瘤组织普鲁士蓝染色:抗 hMAM-USPIO 组较非靶向组细胞内铁染颗粒数量明显增多,初步证明所制备抗 hMAM-USPIO 的乳腺癌靶向造影能力。结论采用共价键法制备的乳腺癌靶向抗hMAM-USPIO 在荷瘤动物模型 MRI 及肿瘤组织普鲁士蓝染色中均证实具备一定靶向能力。Objective To synthesize a breast cancer targeted ultra-small super paramagnetic iron oxide(USPIO)and verify its targeting efficiency in vivo by comparing with histopathology.Methods USPIO was synthesized by coprecipitation chemical method. Monoclonal antibody of mammaglobin,specially expressed by breast cancer,was conjugated to USPIO by chemical covalent bond to synthesize targeted USPIO and its average diameter,content of Fe were determined.The differences of SI in tumorbearing animal model and Fe mark rate in tumor cells between non-targeted USPIO and anti-hMAM USPIO were studied.Results The average diameter of anti-hMAM USPIO was 50.37±13.10 nm and its content of Fe was 20.18 mg/mL.MR in tumor-bearing animal model:The difference of SI decrease after injection was more remarkable in anti-hMAM USPIO than non-targeted USPIO(P 〈0.01).Prussian blue staining of tumor cells:More Fe particles were found in anti-hMAM USPIO group than non-targeted USPIO.Targeting efficiency of anti-hMAM USPIO for breast cancer was preliminarily verified.Conclusion The targeting efficiency of anti-hMAM US-PIO,synthesized by covalent bond method,was verified by both MR scanning in tumor-bearing animal model and Prussian blue staining in tumor cells.
