摘 要:目的 探讨首乌益智胶囊对Notch通路的调控作用.方法 采用永久性双侧颈总动脉结扎法建立慢性脑缺血认知障碍大鼠模型,随机分为首乌益智胶囊组、脑复康组、模型组、假手术组,每组20只大鼠.首乌益智胶囊组大鼠给予首乌益智胶囊(1 ml/100 g),脑复康组给予脑复康(1 ml/100 g),模型组及假手术组给予等量的生理盐水灌胃,共28 d.电镜下观察海马超微结构;实时定量PCR法检测海马Notch1、Hes3及Stat3 mRNA表达.结果 与假手术组比较,模型组、脑复康组和首乌益智胶囊组大鼠海马Notch1、Hes3及Stat3 mRNA表达显著升高(P <0.05 ~0.01).与模型组比较,脑复康组及首乌益智胶囊组大鼠海马Notch1、Hes3及Stat3 mRNA表达显著升高(P <0.05 ~0.01).与脑复康组比较,首乌益智胶囊组大鼠海马Notch1、Hes3及Stat3 mRNA表达显著升高(均P<0.05).结论 首乌益智胶囊可以上调Notch1、Stat3、Hes3基因的表达,从而减轻慢性脑缺血对神经元的损伤,维持神经细胞结构与功能的完整.Objective To investigate the regulatory effect of shouwuyizhi Capsule on Notch pathway. Methods Permanent bilateral common carotid artery ligation method was adopted to establish the rat model with chronic cerebral ischemia cognitive impairment. The rats were randomly divided into Shouwuyizhi capsule group, piracetam group, model group, sham operation group, and there were 20 rats in each group. Shouwuyizhi capsule group was given shouwuyizhi capsule 1 ml/100 g, Piracetam group was given piracetam 1 ml/100 g, the model group and sham operation group were given the same volume of normal saline by gavage, for 28 d. Ultrastructure of hippocampus were observed under electron microscope. The expression of Notchl, Hes3 and Stat3 mRNA in hippocampus were detected by real-time quantitative PCR method. Results Compared with sham operation group, the expression of Notchl, Hes3 and Stat3 of mRNA in hippocampal of model group, piracetam group and Shouwuyizhi capsule group were significantly increased (P 〈 0.05 - 0.01 ). Compared with the model group, the expression of Notchl, Hes3 and Stat3 of mRNA in hippocampal of piracetam group and Shouwuyizhi capsule group were significantly increased ( P 〈 0.05 -0.01 ). Compared with piracetam group, the expression of Notchl, Hes3 and Star3 of mRNA in hippocampal of Shouwuyizhi capsule group were significantly increased (all P 〈 0. 05 ). Conclusion Shouwuyizhi capsule can upregulate the expression of Notchl, Stat3, Hes3 gene, so as to reduce the damage of chronic cerebral ischemia on neurons, maintain cell structure and function of nerve integrity.
