摘 要:目的:探讨卵丘细胞中生长分化因子9(GDF9)mRNA和骨形态蛋白15(BMP15)mRNA的表达水平进行非侵入性评估卵子发育潜能的临床应用价值。方法:收集252例行卵细胞浆内单精子注射(ICSI)助孕治疗的女性患者的卵丘细胞,采用实时定量PCR检测卵丘细胞中GDF9mRNA、BMP15 mRNA表达水平,并分析其与卵子成熟率、受精率和卵裂率的相关性;同时,将患者按照受精情况、有无优质胚胎和有无临床妊娠进行分组,比较不同组间GDF9 mRNA、BMP15mRNA的表达差异。结果:GDF9 mRNA、BMP15 mRNA表达水平与卵子成熟率、正常受精率、卵裂率均呈显著正相关(P<0.001)。GDF9 mRNA、BMP15 mRNA的表达水平在正常受精组显著高于异常受精组(P<0.05),在优质胚胎组显著高于非优质胚胎组(P<0.01),在临床妊娠组也显著高于未妊娠组(P<0.01)。结论:卵丘细胞中GDF9 mRNA和BMP15 mRNA水平与卵子发育潜能有关,可能成为胚胎选择新的分子指标。Objective:To explore whether the expression levels of growth differentiation factor 9 (GDF9) and bone rnorphogenetic protein 15(BMP15)mRNA in cumulus cells can be used as molecular markers for predicting oocyte developmental potential. Methods:Cumulus cells were collected from two hundred and fif- ty-two female patients who underwent the treatment of intracytoplasmic sperm injection (ICSI). The expres- sion levels of GDFg and BMP15 mRNA in cumulus ceils were detected by RT-PCR. The correlations between them and oocyte developmental potential were analyzed. All the 252 patients were grouped into subrgoups according to whether there were fertilization,top quality embryos,or clinical pregnancy,the GDF9 and BMP15 mRNA expressions were compared among the different subgroups. Results:The expression levels of GDFg and BMP15 mRNA were significantly associated with the rate of ooctye maturation, normal fertilization, and cleavage( P 〈 0. 001 ). The expression levels of GDF9 and BMP15 mRNA in normal fertiliaztion group, high-quality embryo group and pregnant group were significantly higher than those in corresponding groups (P〈0. 05). Conclusions:The expression levels of GDF9 and BMP15 mRNA in cumulus cells are associat- ed with oocyte developmental potential, which may be considered as new molecular markers for predicting ooctye develpmental potential.
