摘 要:目的:观察电针"丰隆"穴对高脂血症大鼠腹腔巨噬细胞炎性反应因子表达的影响,探讨电针治疗高脂血症的可能机制。方法:SD大鼠随机分为正常对照组、高脂组、高脂+普通组、高脂+电针组及高脂+普通+电针组,每组8只。采用高脂饮食饲料饲喂法造模,造模28d后,高脂+普通组、高脂+普通+电针组改用普通饲料喂养,高脂+电针组及高脂+普通+电针组电针"丰隆"穴,治疗30min,每天1次,连续治疗28d。治疗结束后,检测各组大鼠血浆总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)含量,流式细胞术及酶联免疫吸附法检测各组大鼠腹腔巨噬细胞中单核趋化因子-1(monocyte chemotactic protein 1,MCP-1)、细胞间黏附分子-1(intercellular adhesion molecule-1,ICAM-1)、白细胞介素-1γ(interleukin-1γ,IL-1γ)的表达。结果:高脂组大鼠血浆TC、LDL-C较正常对照组明显上升(P<0.01);高脂+普通组大鼠血浆TC、LDL-C与高脂组比较明显下降(P<0.01);电针治疗后,高脂+电针组及高脂+普通+电针组大鼠血浆TC、LDL-C明显下降(P<0.01),TG、HDL-C变化不明显(P>0.05)。高脂组大鼠腹腔巨噬细胞内MCP-1、ICAM-1及IL-1γ的表达较正常对照组显著升高(P<0.01);高脂+普通组大鼠腹腔巨噬细胞内MCP-1、ICAM-1及IL-1γ的表达较高脂组明显下降(P<0.05);高脂+普通+电针组大鼠腹腔巨噬细胞内MCP-1、ICAM-1及IL-1γ的表达较高脂+普通组显著下降(P<0.01)。结论:电针"丰隆"穴能够明显下调高脂血症大鼠血脂中TC、LDL-C水平,可能与其下调高脂血症大鼠巨噬细胞中MCP-1、ICAM-1及IL-1γ的表达有关。Objective To observe the effect of electroacupuncture (EA) stimulation of "Fenglong" (ST 40) on celiac inflammatory factors in rats with hyperlipemia (HLP), so as to reveal its mechanism underlying improvement of HLP. Methods A total of 40 SD rats were randomized into normal control, high fat forage, high fat + common forage, high fat + EA, and high fat + common forage+ EA groups, with 8 rats in each group. The HLP model was established by feeding the animals with high fat forage for 28 days. EA (2 mA, 2 Hz/100 Hz) was applied to bilateral ST 40 for 30 min, once daily for 28 days. Contents of plasma total cholesterol (TO), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C), high-density lipoprotein cholesterol (HDL- C) were detected by using an automatic biochemistry analyzer. Intercellular adhesion molecule-1 (ICAM-1), monocyte chemoattractant protein 1 (MCP-1), and interleukin-1γ (IL-1γ) in macrophages of the abdominal cavity were detected using flow cytometry (FCM). Results Compared with the normal control group, the contents of plasma TO and LDL-C, and celiac macrophages, MOP-1, ICAM-1 and IL-ly contents were significantly increased in the high fat forage group and high fat + common forage group (P〈0.01). In comparison with the high fat forage group, contents of plasma TO and LDL-C, and macrophages' MOP-1, ICAM-1and IL-1γ were considerably down-regulated in the high fat+ EA group (P〈0.01). Similarly, the levels of plasma TO and LDL- C, and macrophages' MOP-1, lOAM-1 and IL-1γ were obviously lower in the high fat+ common forage+ EA group than in the high fat+ common forage group (P〈0.01). No significant differences were found in plasma TG and HDL-C levels among the five groups (P〈0.05). Conclusion EA stimulation of "Fenglong" (ST 40) has a role in down-regulating contents of plasma TO and LDL-C and celiac macrophages' MOP-1, lOAM-1 and IL-1γ in the abdominal cavity in hyperlipemia rats,
