摘 要:目的:探讨心肌梗死大鼠心肌VEGF、HIF、eNOS mRNA的表达及安心颗粒的干预作用。方法:将90只清洁级健康雌性SD大鼠随机分为假手术组、模型对照组、麝香保心丸组(80mg·kg-1·d-1)、安心颗粒小剂量组(0.4g·kg-1·d-1)、安心颗粒中剂量组(0.8g·kg-1·d-1)、安心颗粒大剂量组(1.6g·kg-1·d-1),结扎冠状动脉左前降支,制备急性心肌缺血模型。用药组每天上午8时灌胃给药,假手术组和模型对照组予等量生理盐水灌胃(用药剂量分别按动物体表面积换算),每天1次,分笼饲养,自由饮食。6周后处死大鼠,取梗死区心肌组织总mRNA,用RT-PCR的方法检测心肌HIF-1、VEGF、eNOS mRNA的表达。结果:与模型对照组比较,安心颗粒各剂量组与麝香保心丸组心肌组织中HIF-1、VEGF、eNOS mRNA的表达明显升高(P<0.05);麝香保心丸组与安心颗粒各剂量组相比,心肌组织中HIF-1、VEGF、eNOS mRNA的表达无明显差异(P>0.05)。结论:安心颗粒通过诱导心肌HIF-1、VEGF、eNOS mRNA的表达,促进血管的新生,对缺血性心肌的血管重生有着重要作用。Objective:To investigate the myocardial mRNA expression of vascular endothelial growth factor (VEGF), hypoxia - in-ducible factor (HIF), and endothelial nitric oxide synthase (eNOS) in rats with myocardial infarction and the effects of Anxin granuleson their expression. Methods :Ninety clean healthy female Sprague - Dawley rats were randomly divided into sham - operation group,model control group, Shexiang Baoxin pill group ( 80mg· kg-1·d-1 ) , and low - , middle - , and high - dose Anxin granule groups( 0. 4rag· kg-1·d-1 ,0. 8mg- kg-1·d-1 , and 1.6m kg-1·d-1 ). Ligation of the left anterior descending coronary artery was performed toestablish a model of acute myocardial ischemia. The medicated groups received respective treatments by gavage at 8 AM every day, whilethe sham - operation group and model control group were given an equal volume of normal saline by gavage ( the dose was calculated bythe body surface area) once daily;the rats were fed freely in separate cages. These rats were sacrificed six weeks later. Total mRNA ininfarcted myocardium was extracted, and the mRNA expression of HIF - 1, VEGF, and eNOS was measured by RT - PCR. Results :Com-pared with the model control group,the Anxin granule groups and Shexiang Baoxin pill group had significantly increased mRNA expres-sion of HIF - 1, VEGF, and eNOS in the myocardial tissues (P 〈 0.05 ). There were no significant differences in mRNA expression ofHIF - 1, VEGF, and eNOS in myocardial tissues between the Shexiang Baoxin pill group and all Anxin granule groups ( P 〉 O. 05 ) Conclusion: Anxin granules promote angiogenesis by inducing the mRNA expression of HIF - 1, VEGF, and eNOS in myocardial tissuesand thus play an important role in the revascularization of ischemic myocardium.
