摘 要:目的探讨商陆皂甙甲(EsA)对大鼠的体外杀精作用及其机制。方法采用手术方法采集Sprague-Dawley大鼠精子,随机分为EsA组、生理盐水组及壬苯醇醚-9(N-9)组,采用改良Sander-Cramer法,将不同浓度EsA制剂在体外与大鼠精子作用,测定EsA的杀精作用及杀精最低有效浓度;精子尾低渗肿胀(HOS)试验、伊红Y(EY)染色法检测精子膜的完整性和活力,固相法检测精子顶体酶(ACE)活性。结果作用20 s,最低有效质量浓度为2.0 g·L-1的EsA对大鼠精子具有快速杀精作用;与生理盐水组比较,最低有效质量浓度(2.0 g·L-1)的EsA作用20 s后,精子尾肿胀率显著下降(P<0.05),EY染色率显著升高(P<0.05);与生理盐水组比较,EsA各浓度组ACE活性均显著下降(P<0.05),且药物浓度越大,ACE活性降低越明显,组间两两比较差异均有统计学意义(P<0.05)。结论 EsA对大鼠有体外杀精作用,其机制可能与破坏精子膜和降低ACE活性有关。Objective To study the spermicidal effect of esculentoside A (EsA) on rats in vitro and its mechanism. Methods The Sprague-Dawley rats' sperms were collected by operation method, which were randomly divided into EsA group, normal saline (NS) group and nonoxinol-9 (N-9) group. The different concentration EsA acted on rats' sperms in vitro, and the modified Sander-Cramer assayed was used to detect the spermicidal effect and spermicidal minimum effective concentration (MEC). The hypo-osmotic swelling (HOS) test and the eosin Y (EY) staining were used to detect the integrity of sperm membrane and vitality, the solid-phase method was used to detect the acrosomal enzyme (ACE) activity. Results MEC of EsA at 20 s was 2.0 g·L^ - 1. Compared with NS group, the rate of the tails swollen sperm was significantly reduced in EsA groups ( P 〈 0.05 ), the rate of stained sperm was significantly increased in EsA groups (P 〈 0.05 ), the activity of ACE were all significantly reduced in EsA groups (P 〈 0.05 ), and the reducing was associated with drug concentration, the differences were statistically significant by pairwise comparison( P 〈 0.05 ). Conclusion EsA has spermicidal effect on rats in vitro, the spermicidal effect may occur by the disruption of the sperm membrane integrity and the reducing of the ACE activity.
