摘 要:目的研究千佛菌对衰老模型小鼠免疫失衡的调节作用。方法 40只健康昆明小鼠随机分为对照组、模型组及千佛菌大、小剂量组,每组10只。模型组,千佛菌大、小剂量组小鼠按25 mL·kg-1体质量腹腔注射D-半乳糖,每日1次,连续注射6周,制备模型;对照组小鼠腹腔注射等量生理盐水。造模第21天起,千佛菌大、小剂量组每只小鼠分别以1 000和500 g·L-1的千佛菌0.4 mL·d-1灌胃,对照组及模型组小鼠给予等量生理盐水灌胃,连续3周后,检测比较各组小鼠脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)水平及血清γ-干扰素(IFN-γ)、白细胞介素-10(IL-10)水平和IFN-γ/IL-10。结果与对照组比较,模型组小鼠血清IL-10水平升高,IFN-γ水平、IFN-γ/IL-10降低,差异有统计学意义(P<0.01)。与模型组比较,千佛菌大、小剂量组小鼠血清IL-10水平降低,IFN-γ水平、IFN-γ/IL-10升高,差异有统计学意义(P<0.01)。千佛菌大剂量组血清IFN-γ水平、IFN-γ/IL-10高于小剂量组,差异有统计学意义(P<0.05)。与对照组比较,模型组小鼠脑组织SOD水平明显降低,MDA水平明显升高,差异有统计学意义(P<0.01);与模型组比较,千佛菌大、小剂量组小鼠脑组织MDA水平明显下降,SOD水平明显升高,差异有统计学意义(P<0.05)。结论千佛菌对衰老机体具有一定的免疫调节作用。Objective To study the regulatory effect of Grifola frondosa on immune imbalance of aging model mice. Methods Forty healthy Kunming mice were divided into control group, aging model group, large-dose group and low-dose group randomly with 10 mice in each group. The aging mice model was established with D-galactose by intraperitoneal injection for six weeks in aging model group, large-dose group and low-dose group while the same volume physiological saline was used in control group. From the 21 days after the model made, the mice in large-dose group and low-dose group were given 1 000 and 500 g · L^-l Grifola frondosa by intragastric administration with 0.4 mL· d^-1 , while the mice in control group, aging model group using the same volume of physiological saline for three weeks. The content of superoxide dismutase(SOD) and malonal- dehyde (MDA)in brain tissue, the levels of serum interferon-γ(IFN-γ) , interleukin-10 (IL-10) and the IFN-γ/IL-10 were detected and compared. Resldts Compared with the control group ,the IL-IO content in serum of model group increased significantly, while the IFN-γ, IFN-γ/IL-10 recovery ratio in serum and the SOD in brain of model group decreased signifieandy (P 〈 0. 01 ). Compared with the model group ,the IL-10 content in serum decreased significantly while the IFN-γ, IFN-γ/IL-10 recovery ratio in serum increased significandy in the large-dose and low-dose group. Compared with control group, the MDA content in brain tissue of mice decreased significantly while the SOD content increased significantly in the large-dose and low-dose group (P 〈 0. 05 ). Conclusion Grifola frondosa have definite immune regulatory effect on aging body.
