增液润节汤对家兔膝骨关节炎软骨组织学和血清中SOD及关节液中IL-1的影响

摘　　要：目的观察增液润节汤对家兔膝骨关节炎软骨组织学和血清中超氧化物歧化酶(SOD)、关节液中白介素-1(IL-1)含量的影响。方法健康的新西兰大白兔38只,随机分为4组,即正常组(A组)8只、模型组(B组)10只、仙灵骨葆胶囊组(C组)10只、增液润节汤组(D组)10只。除正常组外,其余3组按照关节腔内注射木瓜蛋白酶的方法造模。在治疗前、后分别检测血清SOD和关节液IL-1的含量。并于治疗后行关节腔大体观察及关节软骨组织学的病理检查。结果 1)增液润节汤组SOD水平(92.82±20.35)U/L与模型组(74.24±15.26)U/L比较明显升高(P<0.05),增液润节汤组IL-1水平(125.73±4.27)μg/mL与模型组(180.26±4.59)μg/mL比较明显降低(P<0.05);2)增液润节汤可减轻模型动物关节软骨病理积分。结论增液润节汤可提高血清SOD水平,降低关节液IL-1水平,从而保护关节软骨,减轻炎症对关节软骨的破坏及所产生的疼痛。Objective To observe the effect of Zengye Runjie Decoction on rabbit knee osteoarthritis cartilage tissue and serum superoxide dismutase （SOD）, synovial fluid interleukin-1 （IL-1） content. Methods 38 New Zealand healthy white rabbits were randomly divided into four groups, namely normal group （group A） with eight rabbits, model group （group B） with 10 rabbits, Xianling Gubao Capsules group （group C） with 10 rabbits, Zengye Runjie Decoctions group （D） with 10 rabbits. Except the normal group, molds were made in the rest three groups according to the joint cavity injection of papain. The serum SOD and joint fluid levels of IL-1 were detected before and after treatment respectively. The articular cavity and articular cartilage histological pathology were examined after the treatment. Results 1 ） In Zengye Runjie Decoction group, the SOD level increased significantly （q =1.45, P 〈 0.05） ,and the level of IL-1 was obviously lower compared with model group, （q = 1.24, P 〈 0.05）. 2） Zengye Runjie Decoction can reduce pathological integral of articular cartilage of model animal. Conclusion Zengye Runjie Decoction can increase serum SOD levels and lower levels of IL-1 in synovial fluid so as to protect the articular cartilage and reduce inflammation of articular cartilage damage and pain.
