摘 要:目的研究田基黄石油醚提取物与5-氟尿嘧啶(5-Fu)联用对人肝癌HepG2细胞作用。方法用MTT法检测田基黄石油醚提取物和5-Fu单用和联用对HepG2细胞生长抑制的影响,用DAPI荧光染色法、Annexin-V/PI双染流式细胞术法和琼脂糖凝胶电泳法观察两药对HepG2细胞形态学,诱导凋亡和细胞DNA降解的影响。结果田基黄石油醚提取物及5-Fu均对HepG2细胞有生长抑制作用,且有协同增效作用;DAPI染色结果表明联合给药引起的细胞核凋亡现象较单独给药组更明显;Annexin-V/PI双染流式细胞术法和琼脂糖凝胶电泳法结果表明,联合给药诱导凋亡明显,有典型的凋亡"DNA梯形条带"。结论田基黄石油醚提取物与5-Fu联用后可提高HepG2细胞对5-Fu的敏感性,发挥促凋亡增效作用。AIM To study the synergistic effect of petroleum ether extracts from Hypericum japonicum Thunb. combined with 5-Fu on apoptosis of the human hepatoma HepG2. METHODS MTT assay was used for the meas- urement of tumor cell viability inhibition by petroleum ether extracts from Hypericum japonicum Thunb. with or without 5-fluorouracil. Similar tests were performed to study cell morphology, induced cell apoptosis, and DNA degradation by DAPI fluorescent staining, AnnexinV/PI double staining, and agarose gel electrophoresis, respec- tively. RESULTS Both petroleum ether extracts from Hypericurn japonicum Thunb. and 5-Fu could inhibit the growth of HepG2 and their combination had synergistic effect and easy induction to HepG2 cell apoptosis with DNA ladder by above test methods. CONCLUSION Petroleum ether extracts from Hypericum japonicum Thunb. can significantly increase the sensitivity of HepG2 hepatoma cells to 5-Fu and induce their cells apoptosis.
