玉屏风散调控变应性鼻炎大鼠IL-6、TNF-α活性的实验研究

摘　　要：目的探讨玉屏风散(黄芪、白术和防风)对变应性鼻炎大鼠白介素-6(IL-6)和肿瘤坏死因子-α(TNF-α)表达的影响。方法选用健康SD大鼠,采用10%甲苯-2,4-二异氰酸酯橄榄油溶液复制变应性鼻炎大鼠模型,并用玉屏风散进行治疗。对大鼠过敏症状进行评分;股静脉取血分离血浆采用酶联免疫法检测IL-6及TNF-α的水平;收集腹腔肥大细胞,采用蛋白印迹法检测IL-6及TNF-α蛋白的表达水平;采用过SABC免疫组织化学法对各组动物鼻黏膜进行染色,观察药物对IL-6和TNF-α表达的影响。结果与模型组比较,玉屏风散能明显降低模型大鼠的过敏症状积分(P<0.05,P<0.01),外周血中IL-6及TNF-α的水平(P<0.05,P<0.01),腹腔肥大细胞IL-6和TNF-α的表达水平(P<0.05,P<0.01),鼻黏膜中IL-6和TNF-α的表达。结论玉屏风散能有效治疗变应性鼻炎,其机制可能与其抑制IL-6和TNF-α的活性有关。AIM To investigate the effect of Yupingfeng Powder （Astragali Radix, Atractylodis macrocephalae Rhizoma and Saposhnikoviae Radix ） on the expression of interleukin-6 （IL-6） and tumor necrosis factor-alpha （TNF-α） of blood plasma in rats with allergic rhinitis. METHODS Healthy and adult Sprague Dawley rats were stimulated by 10% solution of toluene-2,4-diisocyanate olive oil to induce the models for allergic rhinitis , and then followed the administration of Yupingfeng Powder. The scores of allergic syndrome were evaluted and the con- tents of IL-6 and TNF-α of blood plasma in peripheral blood were tested by ELISA. The expression levels of IL-6 and TNF-α in rats peritoneal mast cells （RPMC） were measured with western blotting technique. The tissues of nasal mucosa were dyed with immunohistochemical staining of Strept Avidin-Biotin Complex （SABC）. The effect of Yupingfeng Powder on the activity of IL-6 and TNF-α was evaluated. RESULTS Compared with the model group, Yupingfeng Powder remarktedly decreased the scores of llergic syndrome of the rats （P 〈 0.05, P 〈 0.01 ）, decreased the content of IL-6 and TNF-α in blood plasma （P 〈0. 05, P 〈0. 01 ） , and inhibited the expression of IL-6 and TNF-α of nasal mucosa and in the RPMC （P 〈 0. 05, P 〈 0. 01 ）. CONCLUSION Yupingfeng Powder can treat the rats with allergic rhinitis related to the downregulation of activity of IL-6 and TNF-α.
