化瘀消癥杀胚中药复方诱导人输卵管妊娠滋养细胞凋亡

摘　　要：目的探讨化瘀消癥杀胚中药复方(丹参、赤芍、桃仁、紫草和天花粉)对人输卵管妊娠滋养细胞增殖、凋亡的影响。方法采用CCK8法检测经中药复方水煎剂处理人输卵管妊娠滋养细胞的增殖情况,采用Hoechst染色法和流式细胞术检测48 h后细胞凋亡情况,设未经复方中药处理的细胞为空白对照组。结果与空白对照组相比,各剂量中药复方对人输卵管妊娠滋养细胞增殖均有明显的抑制作用,呈剂量和时间依赖性。48 h IC50为27.86 mg/mL;中药复方高、中、低剂量组中人输卵管妊娠滋养细胞呈现典型的凋亡形态学改变,其凋亡率分别为(24.57±1.08)%、(18.30±1.18)%和(14.37±2.64)%,与空白对照组(9.57±0.76)%相比明显增加(F=47.88,Ρ=0.00)。结论化瘀消癥杀胚中药复方可能通过抑制人输卵管妊娠滋养细胞增殖,促进细胞凋亡从而达到中止输卵管妊娠的目的。AIM To investigate the effect of compound herbs （ Salviae miltiorrhizae Radix et Rhizoma, Paeon- iae Radix rubra, Persicae Semen, Arnebiae Radix and Trichosanthis Radix） with dissolving stasis and eliminating mass on the proliferation and apoptosis of human tubal pregnancy nourishing ceils. METHODS CCK8 assay was used for human tubal pregnancy nourishing cells treated with compound herbs to measure the proliferation level. Hoechst staining and flow cytometry were adopted to determine the cell apoptosis after the treatment with high, me- dium, and low dose of compound herbs for 48 h, using untreated ones as control. RESULTS The proliferation of human tubal pregnancy nourishing cells was inhibited by compound herbs in a dose- and time-dependent manner with IC50 of 27.86 mg/mL. The cells treated with high, medium, and low dose of compound for 48 h presented the typical apoptosis morphological changes, with apoptosis rates of （24.57 ± 1.08）% , （18.30 ± 1.18）% , and （ 14.37 ± 2.64 ） % , respectively. They were higher than those of the control group （9.57 ± 0.76 ） % （ F = 47.88, P = 0. 00）. CONCLUSION The compound may inhibit the proliferation and promote the apoptosis of human tubal pregnancy nourishing cells to achieve the avoidance of ectopic pregnancy.
