摘 要:目的建立两头尖中竹节香附素A的HPLC-蒸发光散射检测器(ELSD)测定方法。方法两头尖乙醇提取物的分析在Agilent 300 Extend-C18色谱柱(4.6 mm×100 mm,3.5μm)实施,流动相为乙腈-水(0.1%甲酸)(43∶57),体积流量1 mL/min,柱温30℃。结果竹节香附素A在0.309~2.060μg范围内线性关系良好,r=0.999 4,平均回收率为97.9%,RSD为1.4%。12个不同产地的两头尖中竹节香附素A含有量为0.11%~0.34%之间,多数地区所产药材能够达到药典要求。结论建立了两头尖中竹节香附素A的ELSD检测方法,它的定量限较UV降低了10倍,可推荐作为定量测定方法。AIM To establish a method of determining raddeanin A from Anemone raddeana Rhizoma by HPLC- ELSD. METHODS The analysis of ethanolic extract of A. raddeana Rhizoma was performed on Agilent 300 Extend-Cls Column (4.6 mm× 100 mm, 3.5 μm) of HPLC- ELSD with acetonitrile-water (0. 1% formic acid) (43 : 57) as mobile phase, 1 mL/min of flow rate and 30 ℃ of column temperature. RESULTS There was a good linear relationship in the range of 0. 309 - 2. 060 μg for raddennin A ( r = 0. 999 4), the average recov- ery was 97.9% , and RSD was l. 4%. Raddeanin A from twelve growing areas fell within 0. 11% 4). 34% , most of which came up to China' s pharmacopoeia. CONCLUSION The limit test of established ELSD method for rad- deanin A from A. raddeana Rhizoma is ten times less than that of UV, and it is recommended as the quantitative standard for this herb.
