摘 要:以同时携带ASPV、ACLSV、ASGV 3种潜隐性病毒的"八棱海棠"、"小金海棠"、"华红"和"JAZZ"组培苗为试材,分别采用2种变温热处理方式结合茎尖培养的脱毒方法,运用RTPCR技术对组培苗内病毒进行跟踪检测,研究了不同苹果品种和砧木对变温热处理结合茎尖培养脱除苹果潜隐性病毒的反应以及脱毒苗的适宜检测时机。结果表明:变温热处理条件不同,病毒脱除效果不同。同一热处理条件下,不同苹果基因型其病毒脱除率也不同。"小金海棠"脱毒率最高,"JAZZ"和"八棱海棠"次之,"华红"最低。同时,不同病毒种类的被脱除率存在差异,ASPV最容易脱除,ACLSV居中,ASGV最难脱除。脱毒完成后180d,病毒总检出率最高;220d,3种病毒均有检出,检出顺序为ASGV最先,ACLSV次之,ASPV最晚。因此,对脱毒苗进行多次跟踪检测十分必要。Taking four apple rootstocks and cultivars(‘Malus robusta Rehd.’,‘Malus xiaojinensis.’,‘Huahong’,‘JAZZ’)infected with ASPV,ACLSV and ASGV as materials,alternating heat therapies in combination with shoot tip culture were used to eliminate apple viruses,the RT-PCR system was used to detect and real-time track viruses in virusfree plantlets,the responses to alternating heat therapy in combination with shoot tip culture on latent virus elimination and suitable detection time in different cultivars and rootstocks vitro were studied.The results showed that under different heat treatment conditions,rates of virus-elimination were different.Rates of virus-elimination under same treatment differed from plant to plant.‘Malus xiaojinensis’ listed the top followed by‘JAZZ’and‘Malus robusta Rehd.’,while‘Huahong’ranked number three.However,viruses were always inactivated in the same order:first ASPV,followed by ACLSV,last ASGV.Additionally,among the four detections,the detection rate of ASGV was highest.About 180 days,the detection rate of all latent virus reached apeak.Until 220 days,ASGV,ASPV and ACLSV were detected completely.And the detection sequence was that ASGV was detected first,ACLSV followed,ASPV was the latest.So it was necessary to examine more than once in apple virus-free seedlings.
