摘  要:目的:探讨桉叶油对维甲酸(RA)致畸胎大鼠四肢骨骼发育的影响。方法 SD 孕大鼠42只,随机分为6组,每组7只,正常对照组、RA 组、溶剂对照组(花生油+ RA)及桉叶油100,200和300 mg·kg-1+RA 组。溶剂对照组于孕第7~14天灌胃给予花生油,每只2 mL,每天1次,孕第10天灌胃1次 RA 40 mg·kg-1。桉叶油3个剂量组于孕第7~14天分别灌胃给予桉叶油,每天1次,孕第10天灌胃1次RA 40 mg·kg-1。RA 组于孕第10天灌胃1次 RA 40 mg·kg-1。各组于孕21 d 处死孕鼠取胚胎,测量胎鼠体质重、身长和尾长。每窝活胎随机提取1/2~1/3胎鼠用阿利新蓝和茜素红进行骨骼染色,体视镜下观察骨骼发育,测量四肢长骨长度和骨化长度。结果 RA 灌胃各组胎鼠的体质量、身长和尾长均明显低于正常对照组(P﹤0.05,P﹤0.01);桉叶油各组胎鼠体质量和身长明显高于 RA 组(P﹤0.05,P﹤0.01);溶剂对照组胎鼠体质量、身长和尾长与 RA 组比较无差异;桉叶油各组胎鼠尾长与 RA 组比较无明显变化。RA 各组胎鼠四肢骨的骨长度和骨化长度均低于正常对照组(P﹤0.05,P﹤0.01);桉叶油各组四肢骨骨长度均高于RA 组和溶剂对照组(P﹤0.05,P﹤0.01),溶剂对照组四肢骨骨长度与 RA 组比较无差异;桉叶油各组四肢骨骨化长度高于 RA 组和溶剂对照组(P﹤0.05,P﹤0.01),溶剂对照组四肢骨骨化长度与 RA 组比较无差异。结论桉叶油对 RA 引起胎鼠四肢骨骨骼发育迟缓和骨化迟缓有一定的拮抗作用。OBJECTlVE To study the effect of eucalyptus oil on fetus skeletal development of retin-oic acid( RA)induced teratogenic rats. METHODS Forty-two pregnant SD rats were randomIy and equalIy divided into 6 groups:eucalyptus oil 100,200 and 300 mg·kg-1+RA groups,solvent group(peanut oil+RA)group,RA group and normal group with normal diet. Three eucalyptus oil and solvent groups were ig given eucalyptus oil in 100,200,and 300 mg·kg-1 and peanut oil 2 mL at the 7th to 14th day of gestation,respectively,and RA 40 mg·kg-1 at the 10th day of gestation. RA group were ig given RA 40 mg·kg-1 at the 10th day of gestation. All pregnant rats were sacrificed at the 21th day. The Iength of"nbsp;fetus body and tail as welI as body mass of the embryos were measured. 1/ 2-1/ 3 fetal rats from each Iitter were randomIy extracted to observe skeletal development under a stereoscope by staining with alcian bIue and alizarin red S. The Iength of Iimbs and bone ossification was measured. RESULTS The fetus body mass,body Iength,tail Iength of normal group significantly surpassed those in the other groups(P﹤0.05,P﹤0.01). The fetus body mass and body Iength of each eucalyptus oil group were significantly better than RA group(P﹤0.05,P﹤0.01). The fetus body mass,body Iength and tail Iength of solvent group were not significantly different from those in the RA group. The fetus tail Iength in eucalyptus oil groups had no significant difference compared with RA group. The Iimb and bone ossification of normal group were Ionger than in RA group(P﹤0.05,P﹤0.01). The Iimb Iength of the eucalyptus oil groups was Ionger than in RA group and solvent control group(P﹤0.05,P﹤0.01),but there was no difference between the solvent control group and RA group. The Iimb ossification of each eucalyptus oil group was also Ionger than that in RA group and solvent control group(P﹤0.05,P﹤0.01),but there was no difference between the solvent control group and RA group.
