摘 要:目的研究炙甘草汤对转化生长因子(TGF-β1)与G-CSF分泌的影响。方法选取炙甘草汤与右归饮2个方剂,脐静脉内皮细胞设空白对照组、炙甘草汤组、右归饮组及混合组,每组分别加入相应中药液,于培养第3,6,9天收集细胞培养上清液,ELISA检测上清液中TGF-β1与G-CSF的分泌水平。结果 TGF-β1分泌水平:给药组均明显刺激脐静脉内皮细胞产生TGF-β1,与空白对照组相比差异有统计学意义(P<0.001)。实验初期(第3,6天)炙甘草汤刺激效果较强,实验后期(第9天)混合给药组表现出更好的刺激效果。G-CSF分泌水平:给药组均明显刺激脐静脉内皮细胞产生G-CSF,与空白对照组相比,P<0.001。混合给药组刺激效果最明显,其次为炙甘草汤组。给药后细胞因子分泌均随时间延长而增加。结论炙甘草汤能增强细胞TGF-β1与G-CSF的分泌,当有右归饮辅助时这种效应更加明显,其刺激增强作用有时间累积效应,随时间延长TGF-β1与G-CSF的分泌增多。Objective To investigate the effect of Prepared Licorice Decoction on TGF-β1 and G-CSF production of umbilical vein endothelial cells. Methods Two different Chinese herbal medicine prescriptions were used in this research. Umbilical vein endothelial cells were divided into four groups (control group, prepared licorice decoction group, youguiyin group and mixed group). The cell culture supematant was collected at the 3 rd,6 th,9 th day after herb liquor added into the cell cul- ture system.TGF-β1 and G-CSF level were detected by ELISA.Results 1)TGF-β1 production:The TGF-β1 level was high- er in all three herb liquor groups than that of control group ( P 〈 0. 001 ). The TGF-β1 level of Prepared Licorice Decoction group was highest than that of other groups at day 3 and day 6, but the TGF-β1 level was highest in mixed group than that of the other at day 9.2)G-CSF production:The level of G-CSF was higher in all three herb liquor groups than that of control group ( P 〈 0. 001 ). The level of G-CSF in mixed group was highest, and then is the level in prepared licorice decoction group. Conclusion The Prepared Licorice Decoction n could stimulate umbilical vein endothelial cells secrete higher level of TGF-β1 and G-CSF than control group, and the stimulatory effect was stronger when mixed with youguiyin. The level of TGF-β1 and G-CSF were growing higher as time goes on.
