摘 要:利用青岛农业大学动物医学实验室研制的4种不同配比关系的丙酮酸乙酯(EP)复方混悬剂,对大肠杆菌感染小鼠进行治疗试验,以得到EP与头孢噻呋(CE)的最佳配比量,选出一个最佳组方,并比较不同的EP剂量、给药时间对治疗结果的影响。试验将340只昆明小鼠随机分17组,每组20只,注射大肠杆菌ATCC 25922菌液后,分别于1、4、8h注射4种EP复方混悬剂(EP分别为40、80、120、160mg/kg)和1种CE单方混悬剂(CE为5mg/kg)。观察各组小鼠7d内的死亡情况并做统计学分析。结果显示,攻毒后1和4h进行治疗,EP为40、80mg/kg的复方混悬剂治疗效果显著优于CE单方混悬剂(P<0.05),但2种EP复方混悬剂间无显著差异(P>0.05)。攻毒后8h进行治疗,注射4种EP复方混悬剂和1种CE单方混悬剂均无显著治疗效果(P>0.05),且与阳性对照组相比差异不显著(P>0.05)。结果表明,在小鼠大肠杆菌攻毒试验中,EP 40和80mg/kg的复方混悬剂治疗效果明显优于CE单方混悬剂,能显著提高小鼠治愈率,2种复方混悬剂间无明显差别,但从更安全、更经济的角度考虑选择低剂量的EP 40mg/kg复方混悬剂更为合适。In order to obtain the optimal therapeutic dosage of ethyl pyruvate (EP) in compound preparation and compare the effects of different concentrations of EP and administration time on treatment outcome,four different mixture ratios of sus- pensions composed by EP that prepared by Veterinary Medicine Laboratory (Qingdao Agricultural University) were applied to the experimental therapy of mice infected with E. coll. 340 Kunming mice were divided into 17 groups randomly (20 in each group) and then were injected with E. coli ATCC 25922. A ceftiofur (CE) single suspension (CE 5 mg/kg) and four kinds of EP compound suspension (EP was 40,80,120 and 160 mg/kg,respectively) were injected into the mice after 1,4 and 8 h in or der to observe the mortality rates of mice and analyze statistically. The results showed that the effect of compound suspension groups of EP 40 and 80 mg/kg were much better than CE single suspension group at 1 and 4 h after vaccination (P〈0.05) ,but there was no significant difference between the two compound suspension groups (P〈0.05). At 8 h after vaccination, the therapeutic effect of all compound suspension groups and single suspension group was not significant and there was no signifi- cant difference compared to positive control group (P:〉0.05). The therapeutic effect of compound suspensions of EP 40 and 80 mg/kg were significantly better than CE single suspension, and could help to remarkably improve the cure rate of mice, but there was no significant difference between the two compound suspension groups. It was recommended to choose compound suspension of EP 40 mg/kg from the perspective of security and economy.
